Lnc RNA FER1L4对神经胶质瘤细胞增殖侵袭和凋亡能力的影响

摘要

目的：研究Lnc RNA在神经胶质瘤细胞中的表达，及其对胶质瘤细胞增殖，侵袭及凋亡能力的影响。方法采用RT-PCR检测Lnc RNA FER1L4在不同胶质瘤细胞株S373-MG，U251，U87-MG，SHG-44，LN-18和对照星形胶质细胞1800中的的表达，SiRNA技术沉默FER1L4表达后，采用CCK-8，Transwell侵袭实验，流式细胞技术检测胶质瘤细胞增值、侵袭、凋亡的变化。结果 Lnc RNA在胶质瘤细胞株中表达明显高于正常对照星形胶质细胞。SiRNA下调Lnc RNA表达后，发现神经胶质瘤细胞增殖与侵袭能力明显下降，凋亡细胞数明显增加。结论 Lnc RNA FER1L4可能在胶质瘤发生与发展过程中发挥了重要作用。%Objective To determine the expression of LncRNA FER1L4 and evaluate its clinical value in glioma. Methods The expression of LncRNA FER1L4 in glioma cell lines S373-MG,U251,U87-MG,SHG-44 and normal astrocyte 1800 was determined by RT-PCR. SiRNA targeting FER1L4 was transfected into S373-MG and U251 glioma cell lines,and cell proliferation,invasion,and apoptosis were examined by CCK-8 assays, transwell chamber assays,and Annexin V/PI analysis,respectively. Results The expression of FER1L4 in glioma cell lines was obviously higher than that in the normal astrocyte. Furthermore,by down-regulating FER1L4 using siRNA,the invasiveness and proliferation of the astrocyte decreased considerably,while the apoptosis increased. Conclusions FER1L4 plays an important role in the occurrence and progression of glioma.
