脂氧素受体激动剂BML-111对慢性神经病理性疼痛大鼠治疗的研究

         

摘要

ObjectiveTo test the effect of BML-111 on neuropathic pain induced by L5/6 spinal nerve ligation.MethodsWistar rats were used for establishment of rats L5/6 spinal nerve ligation model. Mechanical paw withdrawal threshold and thermal paw withdrawal threshold were measured after intrathecal injection of different dose BML-111. Two hoursafter the intrathecal injection of 100ug/kg BML-111,the rats ipsilateral spinal cord lumbar enlargement were acquired and measured for IL-1β,IL-6,TNF-α.ResultsBML-111 dose dependently suppressed neuropathic pain behavior with maximum possible effect(67%)and ED50(16µg/kg)for mechanical stimulation;69% and 22µg/kg for thermal stimulation. 100µg/kg BML-111 reduced IL-1β,IL-6,TNF-α concentration inipsilateral spinal cord(P<0.05).Conclusions BML-111 dose dependently suppressed neuropathic pain behavior,the reason may be attribute to the reduction of the infl ammatory mediators production.%目的:观察脂氧素受体激动剂BML-111对L5/6脊神经结扎术后大鼠的镇痛效果。方法选择Wistar大鼠作为实验动物,建立L5/6脊神经结扎动物模型。观察鞘内给予不同剂量的BML-111对机械痛和热痛的抑制作用。鞘内给予100µg/kg BML-111并检测对患侧脊髓腰膨大部位IL-1β、IL-6、TNF-α含量的影响。结果 BML-111剂量依赖性抑制对L5/6脊神经结扎所导致的神经病理性疼痛;其机械痛和热痛最大抑制率分别是67%和69%;半数有效量分别为16µg/kg和22µg/kg。鞘内给予100µg/kg BML-111可有效降低患侧脊髓腰膨大部位IL-1β,IL-6, TNF-α含量(P<0.05)。结论脂氧素受体激动剂BML-111可有效减轻L5/6脊神经结扎术后大鼠神经病理性疼痛,其机制可能与抑制脊髓背角炎症反应有关。
