响应面法优化亚麻籽粕不溶性膳食纤维提取工艺

         

摘要

[目的]采用响应面法研究亚麻籽粕不溶性膳食纤维的最佳提取条件.[方法]以亚麻籽粕为原料,采用碱性蛋白酶水解.在单因素试验基础上,选取酶解温度、时间、加酶量(质量分数)和料液比为响应变量,以不溶性膳食纤维提取率为响应值,利用Box-Behnken试验设计方案和响应面分析法,建立不溶性膳食纤维提取率与响应变量的回归方程,并确定最佳提取条件.[结果]在提取率的二次多项模型中,温度、时间、加酶量在一次项中表现差异显著,温度、料液比、加酶量在二次项中表现差异显著.[结论]亚麻籽粕不溶性膳食纤维的最佳提取条件为:酶解温度55℃、酶解时间4h、料液比1:20、加酶量9%,此条件下水不溶性膳食纤维得率为52.05%,与预测值52.5%较为一致.%[Objective] The purpose of this study is to evaluate the optimum extraction conditions of insoluble dietary fiber in linseed meal by response surface analysis method. [Method] Linseed meal was hydrolyzed with alkaline protease. On the basis of single factor test, the hydrolysis temperature, time, enzyme amount (mass fraction) and the ratio of solid -to -liquid were selected as response variables, the insoluble dietary fiber extraction rate was regarded as response value, and insoluble dietary fiber extraction rate and response variables regression equation were established by Box - Behnken experimental design and response surface analysis method, and the optimum extraction conditions were determined. [Result] In the quadratic model of the extraction rate, the temperature, time and enzyme amount had significant effect in a linear equation and the temperature, ratio and enzyme amount had significant effect in quadratic term. [ Conclusion] The optimum extraction conditions were hydrolysis temperature 55℃ , enzymatic hydrolysis time was 4h, the ratio of solid to liquid was 1:20 and the enzyme amount was 9%. Under this condition, the water - insoluble dietary fiber yield was 52.05% , which was consistent with the predictive value of 52.51%.
