富硒黄芪提取物对免疫抑制大鼠免疫球蛋白IgG、IgM及IgA的影响

     

摘要

目的:观察富硒黄芪提取物对免疫抑制大鼠免疫球蛋白G(IgG)、免疫球蛋白M(IgM)及免疫球蛋白A(IgA)的影响.方法:将SD大鼠随机分为空白组,模型组,富硒黄芪提取物高、中、低剂量组和普通黄芪提取物组.除空白组外,其余5组均采用腹腔注射环磷酰胺,构建大鼠免疫抑制模型.造模成功后分组进行干预治疗.给药第7天后,各组大鼠禁食8小时,股动脉取血,采用全自动血球计数仪检测外周血白细胞总数、中性粒细胞和淋巴细胞数及其百分比;用酶联免疫法检测血清中免疫球蛋白IgG、IgM及IgA的含量.结果:与空白组比较,模型组大鼠外周血血象各数据显著降低,差异有统计学意义(P<0.05);与模型组比较,各干预组大鼠外周血血象数据有所升高,差异有统计学意义(P<0.05).与空白组比较,模型组大鼠免疫球蛋白表达水平显著降低,差异有统计学意义(P<0.05);与模型组比较,各干预组大鼠免疫球蛋白表达水平有所升高,差异有统计学意义(P<0.05).富硒黄芪药物组与普通黄芪药物组相比较,富硒黄芪药物组改善免疫抑制大鼠免疫功能的作用更为显著,差异有统计学意义(P<0.05).结论:富硒黄芪提取物能显著提高环磷酰胺所致免疫抑制模型大鼠的免疫功能.%Objective: To observe the effects of extract of selenium-enriched Astragalus membranaceus on immunoglobulin IgG, IgM and IgA in immunosuppressive rats. Methods:The rats were randomly divided into the blank group, the model group, the high, middle and low dose groups of extract of selenium-enriched Astragalus membranaceus as well as the common astragalus extract group. Other five groups were injected intraperitoneally with cyclophosphamide to construct the immunosuppressive models except the blank group. After the modeling, the groups were intervened respectively. After seven days of treatment, rats, fasted for eight hours, were taken blood from the femoral artery. And the peripheral white blood cell counts, neutrophil and lymphocyte counts as well as the percentage of neutrophils and lymphocytes were determined by automatic globulimeter; the contents of immunoglobulin IgG, IgM and IgA in serum were detected by enzyme-linked immunosorbent assay. Results:Compared with the blank group, the data of peripheral blood hemogram in the model group decreased significantly, and the difference was statistically significant (P<0.05); Compared with the model group, the peripheral blood hemogram data of the intervention groups increased slightly, and the difference was statistically significant(P<0.05). And compared with the blank group, the immunoglobulin expression level of model rats decreased significantly, the difference was statistically significant (P<0.05);Furthermore, compared with the model group, the immunoglobulin expression level of the intervention group rats increased, the difference was statistically significant (P<0.05). Compared with the common astragalus medicine group, the immune function improvements of the selenium-enriched Astragalus membranaceus medicine group on immunosuppressive rats were more significant, and the difference was significant (P<0.05). Conclusion:Extract of Selenium-enriched Astragalus membranaceus can significantly improve the immune function of immunosuppressive rat model induced by cyclophosphamide.
