As2O3对人宫颈癌细胞放疗增敏的机制探讨

         

摘要

本文探讨了三氧化二砷(As2O3)对人宫颈癌Hela细胞放射增敏的可能作用机制.采用免疫细胞化学法检测细胞处理后P21和Bcl-2蛋白表达情况,结果显示2umol/L的As2O3和2Gy的r-射线均能升高P21蛋白表达和降低Bcl-2蛋白的表达(P<0.05),As2O3与射线联合则变化更明显(P<0.01).说明As2O3对人宫颈癌Hela细胞具有放射增敏作用可能与其上调P21和下调Bcl-2蛋白表达有关.%To investigate the possible mechanism on arsenic trioxide' s increasing radiosensitivity in human cervical carcinoma Hela Cell Line.We use immunohistochemieal technique detected the expression of P21 and Bel-2 protein after the Hela cells were treated.The results showed that P21 protein was increased and Bcl-2 protein was decreased when As2O3 and irradiation were applied to cells alone (P<0.05), and the change were much more obvious when As2O3 and irradiation were applied eombinatively than either one of them was used alone (P<0.01).So we make a conclusion that As2O3 had radiosensitizing effect on Hela cells of Human cervical carcinoma, which may contribute to up-regulating P21 Protein and down-regulating Bcl-2 protein.
