A novel amperometric bienzymatic biosensor for determination of glucose is developed by modifying thionine(TH) film on the electrochemical pretreated glassy carbon electrode ( GCE) surface. Gold nanoparticles (GNPs) .glucose oxidase( GOD)and horseridesh peroxidase( HRP)are immolilized by electrostatic adsorption and Au-S,Au-N covalent bond self-assembly. Nation solution is dropped on the modified electrode to prepare anti-interferential and fixed membrane. The experimental results show that the performance of the biosensor can be improved by the electrochemical polarization glassy carbon electrode in HNO3 solution. A good linear relationship between response current and glucose concentration in the range of 1 ×10-3 -18 ×10-3mol/L,R =0. 991 7,the detection limit is 5 ×10-5mol/L The biosensor has high sensitivity, long-term stability, excellent interference immunity and reusability.%在硝酸溶液中电化学极化处理玻碳电极(GCE),以硫堇(TH)为电子介体,通过金-硫、金-氮共价键作用和静电吸附作用将纳米金(GNPs)、葡萄糖氧化酶(GOD)和辣根过氧化物酶(HRP)修饰于电极上,最后在电极上滴涂Nafion水溶液制备抗干扰膜并固定酶电极,构建了一种新型双酶葡萄糖生物传感器.实验结果表明:电化学极化处理电极提高了生物传感器的性能,传感器响应电流与葡萄糖浓度在1 × 10-3- 18×10-3 mol/L范围内有良好的线性关系,R=0.9917,检出限为5×10-5mol/L.该生物传感器具有良好的灵敏度、抗干扰性和稳定性,可以重复使用.
