大黄素对人白蛋白诱导人肾小管上皮细胞转分化的影响

         

摘要

Objective To explore the effect of emodin on the transdifferentiation of human renal tubular epithelial cells to mesenchymal induced by human albumin. Methods Human kidney proximal tubular epithelial cell line (HK-2) was divided into blank control group, emodin control group, albumin-induce group and emodin-inhibition group. HK-2 cells were cultivated for 0, 12, 24 and 48 hours to observe the morphological changes by an inverted phase contrast microscope. The expressions of α-SMA, E-cadherin and FN mRNA were detected by reverse transcription polymerase chain reaction (RT-PCR). Results The HK-2 cells were round and grew in a cobblestone morphology in blank control group and emodin control group. The cell body gradually became longer and thinner, the intercellular space became larger under the light microscope after 12, 24 and 48 hour treatment in albumin-induce group. There were similar morphological changes after 12, 24 and 48-hour treatment in emodin-inhibition group. There were no significant differences in expression levels of α-SMA, E-cadherin and FN mRNA between blank control group and emodin control group (P > 0.05). The expressions of α-SMA and FN mRNA increased with time after 24 and 48-hour treatment in albumin-induce group and emodin -inhibition group compared with those of blank control group, but the expression of E -cadherin mRNA was gradually decreased. There were significantly lower expressions of α-SMA and FN mRNA after 24 and 48-hour treatment in emodin -inhibition group than those of albumin -induce group (P < 0.05). Conclusion Emodin can inhibit the transdifferentiation of human renal tubular epithelial cells to mesenchymal induced by human albumin, and can slow down the progression of renal interstitial fibrosis.%目的 探讨大黄素对人白蛋白诱导的人肾小管上皮细胞(HK-2)发生转分化的影响.方法 体外培养HK-2细胞,按处理方式分为空白对照组、大黄素对照组、白蛋白诱导组、大黄素干预组4组.分别于Oh、12 h、24 h、48 h用倒置显微镜下观察其细胞形态变化,同时RT-PCR法检测各样本α-SMA、E-cadherin、FN mRNA的表达水平.结果 空白对照组、大黄素对照组HK-2细胞形态为圆形,融合后呈鹅卵石样.白蛋白诱导组12、24及48 h光镜下可见细胞胞体拉长变细,细胞之间间隙增大.大黄干预组12、24、48 h后可见细胞形态改变与白蛋白诱导组相似,但改变程度较小.空白对照组和大黄素对照组表达α-SMA、E-cadherin、FN的mRNA水平差异无统计学意义(均P> 0.05).与空白对照组比较,24和48 h白蛋白诱导组、大黄素干预组α-SMA、FN的mRNA随时间延长表达逐渐增强,E-cadherin mRNA逐渐减弱.24和48 h大黄素干预组上述指标的表达改变弱于白蛋白诱导组(均P< 0.05).结论 大黄素可以抑制人白蛋白诱导的HK-2细胞转分化,延缓肾间质纤维化的进展.
