摘要:ToexplorethestructuralcharacteristicofribosomalproteinS29(rpS29)geneofAiluropodamelanoleuca (giantpanda),primersweredesignedbasedontheknownnucleotidesequenceofrpS29genestoclonethecDNAand genomicsequencesofthisgenefromgiantpandabyRT-PCRandPCRstrategyrespectively,andthentheclonedcDNA andgenomicsequencesweresequencedandanalyzedpreliminarily.TheresultsindicatedthatthecDNAfragmentofthe rpS29fromthegiantpandais205bpinsizeinlength,containinganopenreadingframe(ORF)of171bp,encoding56 aminoacids.Thelengthofthegenomicsequenceis1598bp,withthreeexonsandtwointrons.Thecodingsequence showsahighdegreeofhomologytothoseof Homosapiens,Bostaurus,Rattusnorvegicusand Musmusculus with 75%,94.74%,89.47%and88.89%respectively,Whileallthehomologiesforaminoacidsequencesarehighupto 100%.PrimarystructureanalysisrevealedthatthemolecularweightoftheputativerpS29proteinis6.68KD,witha theoreticalpIof10.63.Basedontopologyprediction,therearethreedistincttypesoffunctionalsitesintherpS29 proteinofgiantpanda.
