转Bt基因抗虫棉外源Bt蛋白的差异表达

         

摘要

[Objective] Studying on the temporal and spatial dynamics of Bt protein expression in Bt transgenic cotton and determining the relation of Bt protein expression and Bt transgenic cotton can effectively reduce the harm of target pests.[Method] In this paper,the Bt protein contents in the different tissues,organs,main stem functional leaves and main stem leaves at different positions of transgenic cotton cuhivar (GK19) in different growth stages were studied.[Result] The Bt protein contents of all tissues and organs were different,which were in order of seedling > flower > boll > bud > root > stem.There was a reduction tendency of Bt protein in main stem functional leaves in different growth stages with the advance of development.The Bt protein contents among main stem leaves at different positions in different growth stages were obviously different,which indicated the gradual decrease from the first to the seventh leaves in seedling and full budding stage and a gradual stabilization after a slow increase from the first to the seventh leaves in full flowering and bolling stages.[Conclusion] The Bt protein in all growth periods,all tissues and organs of Bt transgenic cotton were expressed,which changed with temporal and spatial dynamics.%[目的]研究转Bt基因抗虫棉外源Bt蛋白表达的时空规律,明确Bt蛋白表达量和转Bt基因抗虫棉抗性的内在联系,更加有效降低靶标害虫危害.[方法]采用ELISA (Enzyme-linked immunosorbent assay)蛋白定量检测方法,以转Bt基因抗虫棉鄂抗虫棉1号(GK19)为研究材料,对转Bt基因抗虫棉不同组织器官、不同生长时期主茎功能叶和不同生长时期主茎不同叶位叶片的Bt蛋白含量进行检测.[结果]转Bt基因抗虫棉不同组织器官Bt蛋白含量存在差异,苗期叶片最高,花、蕾、铃次之,根、茎较低;不同生长时期主茎功能叶Bt蛋白含量呈现出随生长发育时间推进而降低的趋势;不同生长时期主茎不同叶位叶片Bt蛋白含量差异较大,苗期和盛蕾期第1~7叶呈现出逐渐下降的趋势,而盛花期和盛铃期第1~7叶呈现出先缓慢升高后逐渐稳定的趋势.[结论]Bt蛋白在转Bt基因抗虫棉各生长时期,各组织器官都有表达,且表达量呈时空动态变化.
