IL-2对COPD患者CD8+T细胞频率及功能的影响

         

摘要

目的 探讨IL-2在慢性阻塞性肺疾病(COPD)中的免疫调节作用.方法 采集45例COPD急性发作期患者、33例COPD缓解期患者以及25例健康人外周血,采用ELISA检测血清中IFN-γ、IL-2浓度.提取PBMC后分别设置IL-2刺激组和未刺激组,多色流式分析技术检测CD8+T细胞频率,胞内细胞因子染色技术检测CD8+ IFN-γ+T细胞的频率.结果 COPD患者急性发作期IL-2、IFN-γ浓度较临床缓解期和对照组显著增高(P均<0.01);与未刺激组相比,IL-2刺激组CD8+T细胞频率及分泌IFN-γ显著增加(P<0.05).结论 IL-2可通过对CD8+T细胞的免疫调节效应参与COPD的发生发展过程.%Objective To investigate the immunoregulatory role of IL-2 in the chronic obstructive pulmonary disease ( COPD). Methods IL-2 and IFN-γ in serum from 78 patients with COPD and 25 healthy subjects were quantitively analyzed using enzyme-linked immunosorbent assay ( ELISA). The frequency of CD8+ T cells was analyzed by flow cytometry. IFN-γ was analyzed using intracellular cytokine straining. Results The serum concentrations of IL-2 and IFN-γ in acute exacerbations of COPD patients were significantly higher than those in stable and control groups ( all P <0.01). There were higher frequencies of circulating CD8+ T cells and CD8+ IFN-γ + T cells in the IL-2 stimulation group than those of no IL-2 stimulation group (all P<0.05). Conclusons IL-2 play an important role in the genesis and development of COPD by immunoregulating the frequency and function of CD8+ T cells.
