胃腺癌组织中 RhoC、CD105蛋白的表达变化及意义

         

摘要

目的:观察胃腺癌组织中RhoC、CD105蛋白的表达变化,并探讨其意义。方法采用免疫组化SP法检测108例份胃腺癌组织(胃癌组)、40例份癌旁不典型增生组织(癌旁组)、30例份正常胃黏膜组织(正常组) RhoC、CD105蛋白[ CD105蛋白表达以微血管密度( MVD)表示],并分析RhoC、CD105蛋白两者间及其与胃腺癌临床病理参数的相关性。结果胃癌组、癌旁组、正常组RhoC蛋白阳性率分别为86.1%、60.0%、26.7%,胃癌组、癌旁组分别与正常组比较,P均<0.01;MVD分别为32.63±3.79、12.03±1.32、10.76±1.96,胃癌组与癌旁组、正常组比较,P均<0.01。 RhoC蛋白表达与胃腺癌临床分期、浸润程度、淋巴结转移有关,CD105蛋白与胃腺癌细胞分化程度、临床分期、浸润程度、淋巴结转移有关(P均<0.05)。胃腺癌组织中RhoC蛋白表达阳性组MVD高于阴性组。结论高表达的RhoC参与胃腺癌浸润和淋巴结转移,并与胃腺癌组织中微血管生成有关,联合检测RhoC、CD105蛋白有助于胃腺癌的诊断,RhoC蛋白或可作为判断胃癌恶性行为的重要生物学标记物之一。%Objective To investigate the expression changes of RhoC and CD 105 proteins in gastric carcinoma tissues and to study their significance .Methods The expression of RhoC and CD 105 proteins was detected by immunohistochemis-try in the normal gastric mucosa tissues (n=30), gastric hyperplasia tissues (n=40) and gastric carcinoma tissues (n=108).The correlations between RhoC and CD 105 protein[microvessel density (MVD)], and with the clinic pathological parameters were all studied .Results The positive expression rates of RhoC in the groups of normal gastric mucosa , gastric hyperplasia and gastric carcinoma were 26.7%, 60.0%and 86.1%, respectively (all P<0.01).The MVD indexes in the groups of normal gastric mucosa , gastric hyperplasia and gastric carcinoma were 10.76 ±1.96, 12.03 ±1.32 and 32.63 ±3.79 (all P<0.01).The protein expression of RhoC was related with the clinical stages , depth of tumor infiltra-tion and lymph node metastasis of the gastric adenocarcinoma;CD105 was related with the differentiated degree , clinical sta-ges, depth of tumor infiltration and lymph node metastasis of the gastric adenocarcinoma cells (all P<0.05).The MVD was higher in the positive group about the expression of RhoC protein than that in the negative group .Conclusions The high expression of RhoC participates the tumor infiltration and lymph node metastasis of gastric adenocarcinoma tissues , and RhoC is likely to promote angiogenesis in gastric carcinoma .The combined detection of RhoC and CD 105 is helpful to the di-agnosis of gastric adenocarcinoma .RhoC protein may be used as one of the important biological markers in the diagnosis of gastric carcinoma .
