'嗅三针'对阿尔茨海默病小鼠空间记忆能力和海马APP、Aβ蛋白表达的影响

         

摘要

Objective:To observe the intervening effect of"Xiusanzhen"Electroacupuncture(EA)on spatial memory ability and APP、Aβ protein expression in hippocampus of mice with Alzheimer's disease(AD),in order to clarify its therapeutic mechanism based on the olfactory nerve pathway in alleviating AD.Method:40 AD mice were randomly devided into 4 groups:AD model group,"Xiusanzhen"EA group,"Xiusanzhen"EA with olfactory nerve transected group(ONT+EA),Donepezil HCl group,with 10 mice respectively in each group,then took 10 negative mice as normal control group.Electroacupuncture intervention of"Xiusanzhen"was applied to stimulate bilateral LI 20 and EX-HN3,intragastric administration of donepezil HCI in AD mice was performed at the dose of 3 mg/(kg· d),once daily intervention(except weekends)for 4 weeks in all.Morris water maze was applied to test spatial memo-ry ability of mice.Western blot was applied to test APP、Aβ protein expression in hippocampus of mice.Results:"Xiusanzhen"EA group and Donepezil HCl group could increase number of traversing platforms and target quadrant percent of total time,meanwhile,reduce APP、Aβ protein expression in hippocampus of AD mice(P<0.05);ONT+EA group had no significant differences in influencing indexes of AD mice(P>0.05),there were no significant difference between"Xiusanzhen"EA group and donepezil HCL group(P>0.05).Conclusion:"Xiusanzhen"EA has the effect in ameliorating spatial memory ability and reducing APP、Aβ protein expression in hippocampus of mice with AD,the integrity of the olfactory pathway is the core mechanism of the intervention effect.%目的:观察"嗅三针"对阿尔茨海默病(AD)小鼠学习和海马A PP、Aβ蛋白表达的干预效应,以阐明其基于嗅觉通路治疗AD的作用机制.方法:将40只AD小鼠随机分为AD模型组、嗅三针组、嗅三针加嗅神经切断组、盐酸多奈哌齐组,每组10只,另取10只同窝的阴性小鼠作为正常对照组.通过"嗅三针"对印堂穴和双侧迎香穴行电针刺激,以3 mg/(kg·d)的剂量对盐酸多奈哌齐组AD小鼠进行灌胃,每周干预5次,间歇2 d,共干预4周.小鼠空间记忆能力采用Mor-ris 水迷宫进行检测,海马APP、Aβ蛋白表达采用western blot法进行检测.结果:嗅三针刺激和盐酸多奈哌齐灌胃均能显著增加AD小鼠穿越平台次数和靶象限占总时间百分比,并能显著降低海马A PP、Aβ蛋白表达(P<0.05);嗅三针加嗅神经切断组对各指标表达均无明显影响(P>0. 05),嗅三针组与盐酸多奈哌齐组比较无显著性差异(P>0.05).结论:"嗅三针"具有改善AD小鼠空间记忆能力,降低海马A PP、Aβ蛋白表达的作用,嗅觉通路的完整性是其发挥干预效应的核心机制.
