以紫斑牡丹(Paeonia rockii)种子为研究对象,通过浸种、剥除种皮、纵切、横切、剥出种胚等预处理,进行四唑溶液染色,探究四唑染色法测定牡丹种子生活力的最佳方案.结果表明:用始温50℃水浸种24 h,剥除种皮,放入15℃恒温培养箱培养24h,然后剥出种胚用0.5%的TTC染液染色6h为最佳方案.%Taking the purple peony seed as the research materials,to explore the best solution of the determination of the seed viability,with TTC method by comparison and analysis.This study first designed 14 set of preliminary experiments by a lot of processing like seed soaking,removal of seed skin,longitudinal cutting of embryo,crosscutting of embryo,stripping out embryo,then sorting the analysis of the test results,show that the best solution is first immersed seeds with water in the beginning temperature of 50 ℃ water with 24 h,removing seed skin and putting seeds into constant temperature incubator of 15 ℃ for 24 h,then peeling out of embryo and put them into TTC of 0.5% concentration dyeing 6 h.And then,on the basis of the scheme,to deepen the experiment,the study try to find the best dyeing process when the TTC solution concentration was 0.1%,0.5% and 0.5% when dyeing 1 h or 3 h or 6 h or 9 h or 15 h.The test results show that when the concentration of TTC is 0.5%,dyeing time is 6 h and at the request of the cost savings can obtain best dyeing effects.
