建立比较南苍术和北苍术生品和炮制品的HPLC特征图谱分析方法,同时测定炮制前后苍术素的含量.采用Tiger-kin ODS-3色谱柱(4.6 mm×250 mm,5 μm),流动相为乙腈-水梯度洗脱法,检测波长254 nm,流速1.0 mL·min<'-1>,柱温35℃.南苍术和北苍术特征图谱差异较大;南苍术和北苍术炮制前后成分含量均发生一些变化,其中炮制后苍术素的含量均明显降低.采用特征图谱结合指标性成分含量测定的方法可以较好地控制药材及饮片的质量.该法精密度高、重现性好,所测组分均达到基线分离,可以用于炮制前后南苍术和北苍术的质量评价.%To establish an HPLC specific chromatogram for determination of atractylodin and compare the contents of atractylodin between crude and processed Atractylodes lancea and Atractylodes chinensis. The HPLC separation was performed on a Tigerkin ODS-3 column(4. 6 mm x 250 mm, 5μm) by gradient elution with a mixture of acetonitrile-water as a mobile phase, the flow rate was 1. 0 mL . min-1, and the column temperature was 35℃.The detection wavelength was set at 254 nm. There were difference of characteristic peaks and its relative peak area of HPLC specific chromatograms between between Atractylodes lancea and Atractylodes chinensis. The content of atractylodin decreased significantly in processed Atractylodes lancea and Atractylodes chinensis. Specific chromatograms combined with marker compounds determination can be used for quality control of Chinese medicinal materials and Chinese herbal pieces. This method shows high precision, good repeatability and contents are separation. It can be used for assessment the quality of crude and processed Atractylodes lancea and Atractylodes chinensis.
