研究Ⅲ型1,4,5三磷酸肌醇受体(IP3R-Ⅲ型)在大鼠胰岛β细胞胰岛素分泌颗粒上的定位.利用免疫电镜技术,5nm胶体金标记IP3R-Ⅲ型抗原后定位观察其分布情况.实验表明在胰岛β细胞中,金颗粒大量定位于胰岛素分泌颗粒的内部及颗粒膜上.在分泌颗粒表面的切线部位,也有金颗粒的聚集.本研究证明胰岛素分泌颗粒存在IP3R-Ⅲ型,提示IP3-IP3R3型可能参与了β细胞胰岛素分泌的调节.%To identify the localization of IP3R-HI in secretory granules of rat pancreatic, β cells. 5 nm colloidal gold conjugated IP3 R- HI antibody was used to localize the distribution by immunoelectron microscopy (IEM) . The results show that immunogold particles are detected in the granular core and at the periphery of the secretory granules. In a tangential section across the surface of the granule, there are also clusters of gold particles on the surface of the granules. The localization of IP3R-IH in secretory granules of rat pancreatic β cells suggests that IP3-IP3R3 may mediate insulin secretion.
