载酮洛芬单壁碳纳米管的体外释药及机制研究

         

摘要

用壳聚糖修饰单壁碳纳米管,以酮洛芬为模型药物,制备壳聚糖-酮洛芬-碳纳米管复合物,正交实验优化其处方及制备工艺,红外光谱法(FTIR)和差示扫描量热法(DSC)对其进行表征,用透析法进行体外释药,评价缓释效果,阐明释放机制.结果表明,载药复合物最佳处方工艺为 m(单壁碳纳米管): m(酮洛芬)=1:3,超声功率360 W,超声时间5 min,单壁碳纳米管经壳聚糖修饰后,水溶性和分散性明显改善,载药复合物的载药量可达20.67%,药物表现出明显的缓释效果,释放过程符合一级动力学.%The chitosan-Ketoprofen-carbon nanotube composite was made with chitosan modified sin-gle-walled carbon nanotube as a carrier and Ketoprofen as a model drug.The formulation and prepara-tion was optimized through the orthogonal experiment.Infrared spectroscopy (FTIR)and differential scanning calorimetry (DSC)were used to characterize the product,and the dialysis method was used to study the release property and mechanism.T he results show ed that the optimal prescription of drug-loaded composite was as follows:m(single-walled carbon nanotube): m(Ketoprofen)=1:3,the ul-trasonic power was 360 W and the ultrasonic time was 5 min.The water solubility and dispersion of single-walled carbon nanotube were significantly improved after modified with chitosan.The drug load capacity of the composite could reach a maximum of 20.67%.The drug showed a sustained-release effect and its process was in accordance with first-order kinetics.
