姜黄素与卡铂联合用药对鼠卵巢癌转基因细胞系的影响

     

摘要

目的 探讨姜黄素与卡铂联合用药对鼠卵巢癌转基因细胞系T1、T2、T3的影响.方法 采用MTT法检测卡铂、姜黄素单独及联合用药对鼠卵巢癌转基因细胞系增殖的影响,RT-PCR及Western blot检测各组细胞中K-RAS、PI3K、AKT的表达水平.结果 通过MTT法检测细胞增殖抑制率,各浓度卡铂单独作用于鼠卵巢癌转基因细胞系T1、T2、T3的增殖抑制率<30%,与姜黄素联用后,增殖抑制率均>30% (P<0.05),对转染了AKT的T2、T3作用更明显.联合组T1、T2、T3细胞的K-RAS、PI3K (p-PI3K)、AKT (p-AKT)的mRNA及蛋白表达量低于卡铂单用组.结论 鼠卵巢癌转基因细胞系T1、T2、T3对卡铂具有耐药性.姜黄素可逆转其对卡铂的耐药性,且对含有AKT的T2、T3作用更加明显.姜黄素的逆转作用可能与抑制K-RAS、PI3K、AKT的表达有关,其也可能直接抑制PI3K/AKT信号通路.%Objective To investigate the effect of curcumin combined with carboplatin on rat′s ovarian cancer transgenic cell line T1,T2 and T3.Methods The effect of carboplatin and curcumin alone and in combination on proliferation of rat′s ovarian cancer transgenic cell lines was detected by MTT method,and the expression levels of K-RAS,PI3K and AKT were detected by RT-PCR and Western blotting.Results The cell inhibition rate of proliferation of different concentrations of carboplatin alone on rat ovarian cancer transgenic cell line T1,T2 and T3 was less than 30%;the rate was more than 30% after combination use of carboplatin and curcumin (P<0.05);the effect on T2 and T3 which was transfected with AKT was more obvious.The expression of K-RAS,PI3K (p-PI3K) and AKT (p-AKT) in T1,T2 and T3 of combination group was lower than that of carboplatin group.Conclusion Rat ovarian cancer transgenic cell lines T1,T2 and T3 are resistant to carboplatin.Curcumin can reverse the drug resistance to carboplatin,especially for T2 and T3 cells which contain AKT.The reverse effect of curcumin may be related to the inhibition of the expression of K-RAS,PI3K and AKT or direct inhibition of PI3K/AKT signaling pathway.
