P SA、TNF-α及Bax-α在前列腺癌中的表达

         

摘要

目的探讨前列腺特异性抗原( PSA)、肿瘤坏死因子α( TNF⁃α)及B细胞淋巴瘤基因⁃2相关x蛋白( Bax⁃α)与前列腺癌的关系。方法选取前列腺癌患者30例(Ⅰ期12例、Ⅱ期8例、Ⅲ期5例、Ⅳ期5例),正常对照20例,采用电化学发光仪检测患者空腹PSA,放射免疫法测定TNF⁃α,免疫组织化学SP法检测Bax⁃α蛋白的表达,分析Bax⁃α的表达与前列腺癌组织临床病理学特征的关系。结果前列腺癌中Bax⁃α的阳性率高于正常组织( P<0�01),前列腺癌患者的临床分期越高、有淋巴结转移者Bax⁃α的阳性率越高( P<0�01);Ⅰ期、Ⅱ期、Ⅲ期和Ⅳ期患者PSA水平以及Ⅰ期、Ⅱ期、Ⅲ期和Ⅳ期患者的TNF⁃α水平均明显高于正常对照者。结论 Bax⁃α在细胞凋亡调节中起着重要的作用,其过度表达在前列腺癌的发生、发展中扮演着重要的角色。联合检测Bax⁃α、PSA和TNF⁃α水平的变化对前列腺癌的诊断、治疗和预后判断均具有一定的临床价值。%Objective To explore the relationship between prostatic specific antigen( PSA) ,tumor necrosis factor⁃α( TNF⁃α) ,Bax⁃α and prostate cancer. Methods The serum PSA levels were detected by electro⁃chemiluminescence, TNF⁃α examined by RIA, and immunohisto⁃chemical method was used to detect the expression of Bax⁃α in the prostate cancer tissue of 30 cases and 20 healthy persons. Results The expression of Bax⁃α in the patients with prostate cancer was significantly higher than that in healthy persons( P<0�01) . The expression of Bax⁃αwas related with clinical stage,me⁃tastasis in lymph node in prostatic patients (P<0�01).The concentration of TNF⁃α in stage Ⅰ、Ⅱ、Ⅲ and Ⅳ and PSA in stage Ⅰ、Ⅱ、ⅢandⅣpatients was significantly higher than that in control group( P<0�05 or P<0�01) . Conclusions Bax⁃α is correlated with the modulation of proliferation in the development of prostate cancer. The over⁃expression of Bax⁃αplays an important role in the development of prostate cancer. Detection of serum PSA,TNF⁃αand Bax⁃αlevel shows clini⁃cal value for diagnosis,treatment and prognosis prediction of prostate cancer.
