目的 建立化疗药物丝裂霉素、5-氟尿嘧啶诱发小鼠血小板减少、白细胞减少的动物模型.方法 采用不同用法用量的丝裂霉素、5-氟尿嘧啶给予小鼠,在不同时间进行血液学检查,确定丝裂霉素、5-氟尿嘧啶能够引起小鼠血小板减少、白细胞减少的合适剂量和成模时间.结果 腹腔注射丝裂霉素首剂量为25或50 mg·kg-1、维持量为12.5 mg·kg-1,5-氟尿嘧啶腹腔注射首剂量为125或187.5 mg·kg-1、维持量为62.5 mg·kg-1,外周血WBC和PLT均显著降低,与空白组比较,差异有统计学意义(P<0.01).丝裂霉素首剂量50 mg·kg-1、维持量12.5 mg·kg-1,5-氟尿嘧啶首剂量187.5 mg·kg-1、维持量62.5 mg·kg-1,其RBC均降低,与空白组比较,差异有统计学意义(P<0.01).结论 采用上述剂量的丝裂霉素、5-氟尿嘧啶可诱发复制小鼠白细胞减少、血小板减少模型,成功率较高,动物死亡率低.%Objective To establish the animal models of thrombocytopenia, leukopenia induced by mitomycin or 5-fluorouracil in mice. Method The different usages and dosages of mitomycin or 5-fluorouracil were investigated. The amount of platelets, blood cells of the mouse were detected at different times in order to determine the appropriate usage and dosage that can induce thrombocytopenia and leukopenia. Results When mitomycin ip 25 or 50 mg ? Kg-1 as first-dosage and 12. 5 mg ? Kg-1 as maintenance-dosage were injected to the mouse,the amount of platelets and white blood cells were lower than that of control group (P <0. 01). The same effect first-dosage and maintenance-dosage of 5-fluorouracil was 125 or 187. 5 mg ? Kg-1 and 62. 5 mg ? Kg-1. When first-dosage and maintenance-dosage of mitomycin or 5-fluorouracil were added to 50 mg ? Kg-1 and 12. 5 mg ? Kg-1 or 187. 5 mg ? Kg-1 and 62. 5 mg ? Kg-1 ,the amount of red blood cells was lower than that of control group ( P <0. 01). Conclusion Appropriate first-dosage and maintenance-dosage of mitomycin is ip 25 or 50 mg ? Kg-1 and 12. 5 mg ? Kg-1 ,5-fluorouracil is 125 or 187. 5 mg ? Kg-1 and 62. 5 mg ? Kg-1. This usages and dosages can successfully establish the animal models of thrombocytopenia or leukopenia in mice.
