丹酚酸A对过氧化氢诱导的人脐静脉内皮细胞损伤的保护作用

         

摘要

Objective To study the protective effect of salvianolic acid A against the damage caused by hydrogen peroxide (H2 O2 ) in human umbilical vascular endothelial cells(HUVECs) .Methods The oxidative injury model established with H2O2 was used to study the protective effects of salvianolic acid A (10 ,20 ,40 μg · mL -1 ) on the oxidative damage of HUVECs .The activities of SOD ,the level of MDA and the content of LDH as well as NO were measured by spectrophotometric methods as described in the assay kits .The relative caspase-3 activity was detected with colorimetry .Results The results indicated that salvianolic acid A could inhibit the H2 O2-induced reduction of cell activities ,and reduce the releases of MDA and LDH accompanied by the increase in the content of NO and the activities of SOD as well as caspase-3 .Conclusion Salvianolic acid A could prevent HUVECs from oxidative damage .%目的:研究丹酚酸A对人脐静脉内皮细胞(HUVECs)氧化损伤的保护作用。方法采用过氧化氢(H2 O2)诱导的 HU-VECs氧化损伤模型,观察丹酚酸A(10,20和40μg · mL -1)对血管内皮细胞损伤的保护作用,MTT法检测细胞存活率,试剂盒检测细胞上清液中的超氧化物歧化酶(SOD)、丙二醛(MDA)、乳酸脱氢酶(LDH)、一氧化氮(NO),比色法测定Caspase-3的活性。结果丹酚酸A可抑制 H2 O2诱导的血管内皮细胞的凋亡,同时增加SOD的活性,降低M DA的水平,减少LD H的漏出量,增加NO的释放,降低Caspase-3的活性。且丹酚酸A的上述作用均呈质量浓度依赖性。结论丹酚酸A可以有效保护内皮细胞免受氧化损伤。
