金属硫蛋白和瑞舒伐他汀对APOE基因缺失小鼠主动脉核因子-κB表达的影响

         

摘要

目的 探讨金属硫蛋白和调脂药物瑞舒伐他汀对APOE基因缺失小鼠血脂及主动脉核因子-κB表达的影响.方法 高脂饮食喂养并药物处理APOE基因缺失小鼠13周,摘眼球取血检测血脂、肝肾功能及肌酸磷酸激酶等生化指标,处死小鼠后取主动脉,观察主动脉形态学变化和血管壁斑块组织变化,定性分析血管核因子-κB表达强度的变化.结果 与高脂模型组比较,瑞舒伐他汀组血脂水平显著降低(P均<0.05),金属硫蛋白组血脂水平无明显变化(P均>0.05).金属硫蛋白和瑞舒伐他汀组肝肾功能和肌酸磷酸激酶与模型组比较无显著性差异(P均>0.05).瑞舒伐他汀和金属硫蛋白组主动脉内膜厚度和血管壁斑块组织免疫着色较高脂模型组明显减轻(P均<0.05).结论 瑞舒伐他汀降低血脂作用明显,金属硫蛋白对血脂无明显影响,2者均可减轻动脉粥样硬化病变.金属硫蛋白抗动脉粥样硬化作用与其抑制主动脉核因子-κB表达有关.%Objective It is to approach the influence of metallothionein and lipid lowering drugs rosuvastatin on blood lipids and the expression of arterial nuclear factor - κB in APOE gene deletion mice. Methods The APOE gene deficient mice were fed with high fat diet for 13 weeks. Then the eyehalls were picked to get blood to detected the biochemical parameters such as blood fat, liver and kidney function and creatine kinase. The changes of aortic morphology and vascular wall plaque organizational were observed after killed the mice, and the changes of the arterial nuclear factor - κB expression in vascular were qualitatively analyzed. Results Compared with model group, the levels of blood fat were significantly lower with statistical difference in rosuvastatin group ( P <0. 05 ), and no significantly changes in metallothionein group ( P >0. 05 ); there was no difference in function of liver and renal and creatine kinase in metallothionein group and rosuvastatin group ( P > 0. 05 ); the thickness of aortic intima and vascular wall plaque Immunization color density in rosuvastatin group and metallothionein group were significantly reduced. Conclusion Rosuvastatin can significantly lower blood fat; metallothionein has no effect on blood fat. Rosuvastatin and metallothionein may alleviate atherosclerotic lesions. The role of anti-atherosclerosis of metallothionein is related to its inhibition on the expression of NF - κB in aorta.
