人参皂苷Rh2对人食管癌Eca-109细胞TFPI-2表达的影响

摘要

Objective It is to approach the influence of GS - Rh2 on expression of TFPI - 2 of human esophageal cancer Eca - 109 cells. Methods After treatment with different concentration of GS - Rh2 for 48h, the expression of TFPI - 2 mRNA of Eca - 109 cells was measured with RT - PCR, the expression of TFPI - 2 protein of Eca - 109 cells was measured with western blot and the invasion of Eca - 109 cells was analyzed by Transwell invasion assay. Results Treatment with GS - Rh2 at concentration of 40 and 80 mg/L significantly inhibited the invasion and increased the TFPI - 2 mRNA and protein of Eca -109 cells than control group ( P <0.05 ), GS - Rh2 of 10 mg/L was so, but no significant than control group ( P > 0. 05 ). Conclusion GS - Rh2 can increase the expression of TFPI - 2 mRNA and protein of Eca - 109 cells, and inhibit the invasion of its.%目的 探讨人参皂苷Rh2对人食管癌Eca-109细胞组织因子途径抑制物2(TFPI-2)表达的影响.方法 按人参皂苷Rh2不同浓度设置3个实验组(10,40,80mg/L)和1个对照组,分别采用RT-PCR法检测细胞TFPI-2 mRNA表达水平、Western blot法检测TFPI-2蛋白表达水平以及Transwell小室法检测细胞的侵袭能力等.结果 10,40,80mg/L人参皂苷Rh2均可抑制人食管癌细胞Eca-109的侵袭力并能上调TFPI-2 mRNA及蛋白的表达,且40,80mg/L与对照组比较有显著性差异(P均<0.05).结论 人参皂苷Rh2具有上调人食管癌Eca-109细胞TFPI-2基因表达和抑制其侵袭力的作用.
