血必净注射液对急性百草枯中毒大鼠肺组织中炎性因子表达影响研究

         

摘要

Objective It is to observe the effects of Xuebijing injection on the activity of SOD,the content of MDA and the expression of IL-6,TGF-β1in the lung tissue of paraquat-poisoned rats,thus to explore the mechanism of Xuebijing in the treatment of acute paraquat poisoning. Methods On the base of random number table,108 Wister rats were divided into 3 groups:control group,poisoned group,treatment group,each group had 36 rats. After model establishing successfully the treatment group was given Xuebingjing injection 4 mL/kg by intraperitoneal injection,the other two groups were given the same dose of normal water. On 1st,3rd,7th,14th,21st and 28th day,six rats from each group were executed to detect the activity of SOD,the content of MDA and the expression of IL-6,TGF-β1in the lung tissue. Results In the poisoned group,the activity of SOD in the lung tissue of the rats was decreased,the content of MDA was increased,and the expres-sion of IL-6,TGF-β1was increased,and there was a significant difference compared with the control group at every time point (P<0.05). After the treatment of Xuebijing,the SOD activity increased slightly,MDA content increased and IL-6 and TGF-β1expression decreased in the treatment group,which was statistically significant compared with the poisoned group (P<0.05). Compared with the control group,the activity of SOD (1 d to 14 d)was still lower (P <0.05),the content of MDA (1 d to 14 d)and the expression of IL,TGF-β1were still higher (P<0.05)in the treatment group. Con-clusion Oxygen free radicals and cytokine IL-6,TGF-β1play an important role in lung injury induced by paraquat in rats; Xuebijing can relieve the lung tissue injury of paraquat-poisoned rats by regulating these indexes.%目的 观察血必净注射液对百草枯中毒大鼠肺组织中超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量及白细胞介素 -6(IL-6)、转化生长因子 -β1(TGF-β1)蛋白表达的影响,探讨血必净注射液治疗急性百草枯中毒的机制.方法 采用随机数字表法将108只 Wistar大鼠分成对照组、染毒组、干预组,每组36只.干预组造模后给予血必净注射液4 mL/kg腹腔注射,对照组、染毒组给予等量生理盐水腹腔注射.每组分别在干预后1,3,7,14,21,28 d各处死6只大鼠,检测肺组织中 SOD活力、MDA含量及 IL -6、TGF-β1表达量.结果 染毒组大鼠肺组织中 SOD活力下降,MDA含量升高,IL -6和TGF-β1蛋白表达增强,干预后各时间点与对照组比较差异均有统计学意义(P均 <0.05).干预组干预后各时间点 SOD活力均明显高于染毒组(P均 <0.05),但干预后1 ~14 d SOD活力均明显低于对照组(P均 <0.05);干预后1 ~21 d MDA含量和各时间点 IL-6、TGF-β1表达量均明显低于染毒组(P均 <0.05),但干预后1 ~14 d MDA含量和干预后各时间点 IL -6、TGF -β1表达量均明显高于对照组(P均 <0.05).结论 氧自由基和细胞因子 IL-6、TGF-β1均参与百草枯所致肺损伤,而血必净注射液能够通过调节这些指标表达而减轻染毒大鼠肺组织损伤.
