血管内皮生长因子在急性减压缺氧小鼠肺组织中的表达研究

         

摘要

目的 研究血管内皮生长因子( vascuar endpthelial growth factor,VEGF)在急性减压缺氧小鼠肺组织中的表达及意义. 方法 采用低压低氧动物实验舱分别模拟海拔6000 m和8000 m高原缺氧环境,将BALB/c小鼠随机分为正常对照组、缺氧6、9、12、24、48、72 h组,每组10只. HE染色切片观察缺氧不同时间小鼠肺组织病理学的改变情况,免疫组织化学法检测肺组织VEGF的表达水平差异及其规律. 结果 缺氧各组肺组织肺泡间质薄厚不均,肺泡大小不一,部分肺泡腔内见少量红细胞,血管扩张充血出血,且随海拔的增加病理性状况愈严重;VEGF阳性表达强度半定量评分较正常对照组显著增加(P<0. 01),且随海拔的增加及缺氧时间的延长表达增加. 结论 减压缺氧能造成小鼠肺组织损伤,呈现水肿病理表现,并刺激VEGF分泌,最终加重肺水肿和诱导细胞坏死.%Objective To investigate the expression and significance of vascular endothelial growth factor ( VEGF) in lung tissues of mice under acute depression and hypoxia condition. Methods The hypobaric and mionectic chamber was used to simulate 6000 m and 8000 m high altitude hypoxia environment, and BALB/c mice were randomly divided into normal group (n=10), hypoxia for 6 h, 9 h, 12 h, 24 h, 48 h and 72 h groups (n=10 for each group). The pathological changes of lung tissues in different hypoxia times were observed using HE staining, and VEGF expres-sions and its rule of lung tissues were detected using immunohistochemistry method. Results In hypoxia groups, the tis-sues between pulmonary alveoli were different thickness and sizes, and a small amount of red blood cells was observed in some pulmonary alveoli. The vascular extension, hyperemia and hemorrhage were found, and pathological changes were more serious with increasing altitude. Compared with those in normal group, the positive VEGF expression of intensity semi-quantitative scores were significantly increased in hypoxia groups ( P <0. 01 ) , and VEGF expressions were in-creased with increasing altitude and prolonged hypoxia time. Conclusion The lung tissues may be damaged in mice un-der depression and hypoxia condition, and it can induce edema and stimulate VEGF secretion, which may result in more serious pulmonary edema and cell death.
