维生素E联合银杏叶提取物对局灶性脑缺血再灌注损伤大鼠外周血清氧化相关物质浓度的影响

     

摘要

目的:研究维生素E联合银杏叶提取物对局灶性脑缺血再灌注损伤的保护作用及其机制。方法选取80只清洁级SD大鼠,随机分为观察组和对照组(每组40只),制备局灶性脑缺血再灌注损伤模型,对照组给予银杏叶提取物,观察组给予银杏叶提取物的同时给予维生素E,在缺血再灌注后1、3、6、24h每组选取10只测定外周血清中谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)、活性氧类物质(ROS)、丙二醛(MDA)、血小板活化因子(PAF)浓度的变化。结果观察组大鼠各个时间点外周血清GSH-PX、SOD活性值均高于对照组,外周血清ROS、MDA、PAF均低于对照组(P<0.05)。两组GSH-PX、SOD活性均于再灌注后开始下降,6h达到最低值,24h开始升高,而ROS、MDA、PAF均于再灌注后开始升高,6h达到高峰,24h开始下降。结论维生素E联合银杏叶提取物能通过升高外周血GSH-PX、SOD活性与降低外周血ROS、MDA、PAF水平减轻脑缺血再灌注损伤。%Objective To investigate the protective effect and mechanisms of Vitamin E combined with extract of Ginkgo Biloba leaf on focal cerebral ischemia and reperfusion injury in rats. Methods A total of 80 Sprague Dawley rats were randomly divided into control group (n = 40) and observation group (n = 40) to establish models of focal cerebral ischemia and reperfusion injury. The control group was injected with extract of Ginkgo Biloba leaf, while the observation group was injected with extract of Ginkgo Biloba leaf and Vitamin E. A total of 10 rats were selected in each group after ischemia and reperfusion for 1, 3, 6 and 24 h. The concentration changes of glutathion peroxidase (GSH-PX), superox-ide dismutase (SOD), reactive oxygen species (ROS), malondialdehyde (MDA) and platelet activating factor (PAF) in peripheral serum of rats were detected. Results Compared with those in control group, at all time points, the values of GSH-PX and SOD activities were significantly higher, while the values of ROS, MDA and PAF were significantly lower in observation group (P < 0. 05). In the two groups, the GSH-PX and SOD activities were decreased when the reperfu-sion was started, the lowest values were found 6 h after reperfusion, and the increased values were found after reperfusion for 24 h, while the ROS activity and concentrations of MDA and PAF were increased when the reperfusion was beginning, the highest values were found after reperfusion for 6 h, and the decreased values were found 24 h after reperfusion. Con-clusion Vitamin E combined with extract of Ginkgo Biloba leaf can relieve focal cerebral ischemia and reperfusion injury by increasing the GSH-PX and SOD activities and reducing the ROS activity and MDA and PAF concentrations in periph-eral serum in rats.
