目的 探讨含CpG基序的寡脱氧核苷酸对人肺癌细胞株A549放射敏感性的影响.方法 分别用不同浓度CpG ODN处理A549细胞,经β射线照射后,检测细胞增殖和集落形成情况,ELISA法检测细胞TNF-α、IL-12和INF-γ的分泌水平,Griess检测细胞NO的含量.结果 CpG ODN抑制A549细胞的增殖,并呈量效关系.CpG ODN(10 μg/ml)联合β射线照射,抑制A549细胞的增殖作用最强,且增加了TNF-α、IL-12、INF-γ和NO的分泌.结论 CpG ODN对A549细胞有明显的放射增敏作用,其机制可能与CpG ODN抑制A549细胞增殖、增强TNF-α、IL-12、INF-γ和NO的分泌有关.%Objective To investigate the influence of oligodeoxynucleotide containing CpG motif ( CpG ODN ) on the rnradiosensitivity of human lung cancer cell line A549. Methods A549 cells were exposed to CpG ODN at different concentration ( 1 ,3 ,rn10,30 μg/ml ) and β - ray irradiation ( 5 Gy ). The cell proliferation and colony formation were measured afterward. The secretion levels rnof TNF - α,IL - 12 and INF - γ were detected by ELISA method. The NO level was detected by Griess method. Results CpG ODN rninhibited the proliferation of A549 cells, and presented a dose - effect relationship. CpG ODN ( 10 μg/ml ) combined with β - ray rnirradiation had the best effects on inhibiting the proliferation of cells,and it increased the secretion of TNF - α , IL - 12 , INF - γ and rnNO. Conclusion CpG ODN has an obvious radiosensitizing effect on A549 cells,which may be related to its inhibition of A549 cell rnproliferation and enhancement of the secretion of TNF - α,IL - 12,INF - γ and NO.
