caspase-3、HSP70表达在子痫前期发病机制中的作用

摘要

目的：探讨子痫前期患者胎盘组织中caspase-3、HSP70的表达情况及其作用机制。方法：选择本院住院治疗的子痫前期孕妇45例为研究组，正常孕妇45例为对照组。收集两组的胎盘组织标本，采用免疫组化法检测两组胎盘组织中caspase-3、HSP70的表达。结果：研究组胎盘组织中caspase-3蛋白和HSP70蛋白的阳性表达率分别为62.2%、55.6%，均显著高于对照组的4.4%、11.1%，差异均有统计学意义（χ2=11.65、9.37，P<0.01）。随着研究组孕妇的病情加重，胎盘组织中caspase-3蛋白和HSP70蛋白表达均呈上升趋势，重度子痫前期孕妇胎盘组织中caspase-3蛋白和HSP70蛋白的阳性表达率均明显高于轻度子痫前期孕妇（χ2=9.91、7.93，P<0.01）。研究组中HSP70蛋白的表达和caspase-3蛋白的表达呈负相关（r=0.582，P<0.01）。结论：caspase-3和HSP70的异常表达不仅可能参与了子痫前期形成过程，而且也在子痫前期患者病情进展过程中发挥重要作用，两者之间可能存在相互作用。%Objective:To investigate the expression of caspase-3 and HSP70 in the placenta of patients with preeclampsia and its mechanism.Method:45 preeclampsia pregnant women in our hospital were selected as study group and 45 normal pregnant women as control group.Placental tissue samples were collected,and the expression of caspase-3 and HSP70 in the two groups were detected by immunohistochemical method.Result:The positive expression rates of caspase-3 protein and HSP70 protein in placenta tissue of the study group were 62.2%and 55.6%,respectively,which were significantly higher than 4.4%,11.1%of the control group,the differences were statistically significant(χ2=11.65,9.37,P<0.01).The expression of caspase-3 protein and HSP70 protein in placenta tissue were increased with the increase of the severity of the pregnant women in the study group,the positive expression rates of caspase-3 protein and HSP70 protein in placenta of severe preeclampsia were significantly higher than those in mild preeclampsia(χ2=9.91,7.93,P<0.01).In the study group,the expression of HSP70 protein and the expression of caspase-3 protein were negatively correlated(r=0.582, P<0.01).Conclusion:The abnormal expression of caspase-3 and HSP70 may not only be involved in the formation of preeclampsia,but also play an important role in the progression of the disease in patients with preeclampsia, and there may be interactions between them.
