Ets-1在非小细胞肺癌中的表达及其临床意义

原癌基因Ets-1,与肿瘤的发生、浸润转移、血管生成及预后密切相关.通过采用免疫组化SP法和RT-PCR技术检测非小细胞肺癌(non-small cell lung cancer,NSCLC)组织及癌旁正常组织中Ets -1的表达,并分析与临床病理特征及预后的关系.免疫组化结果显示Ets-1蛋白在NSCLC组织中的阳性率为67％ (65/97),显著高于对应的癌旁正常组织0％ (0/30) (P＜0.001).Ets-1阳性率随着肿瘤T分期的增加、淋巴结转移和临床分期的增加而增加(P＜0.05),而与性别、年龄、吸烟、组织学类型及分化程度等无关(P＞0.05).RT-PCR结果显示Ets-1 mR-NA在20例癌组中和癌旁组中的相对表达强度分别为0.5570±0.0593和0.2965±0.0869(P＜0.001).Kaplan-Meier 生存曲线显示,Ets-1阳性组的生存时间显著低于阴性组(P＜0.05).Cox多因素分析模型显示Ets-1不是NSCLC患者的独立影像因素(P＞0.05).结果表明,Ets-1的表达在NSCLC的浸润和转移中扮演了重要的角色,并对NSCLC患者生存期有一定的影响.%Proto oncogene Ets-l,was correlated with tumorigenesis,invasion,metastasis,angiogenesis and prognosis of tumor.The expression of Ets-l was detected by SP immunohistochemistry and RT-PCR in NSCLC and neighbouring normal lung cancer tissue,and the relationship between expression and clinico-pathologic characteristics and prognosis of NSCLC were analyzed.The positive rate of Ets-l protein in NSCLC tissue was 67% (65/97),which is significantly higher than that in neighbouring normal lung cancer tissue 0%(0/30) (P<0.001),respectively.The expression rate of Ets-l protein was increased with higher T stages,lymph node metastasis and higher clinical stages (P<0.05) of NSCLC,but do not associated with sex,age,smoking,histological classification and differentiation (P>0.05).The relative expression intensity of Ets-1 mRNA in 20 pairs of NSCLC tissues and neighbouring normal cancer tissue were 0.5570 ±0.0593 and 0.2965±0.0869 (P<0.001).The survival time of Ets-l-positive patients was poorer than that of Ets-1-negative patients (P<0.05),which was showed by Kaplan-Meier survival analysis.Ets-l expression was not an independent prognostic factor of NSCLC (P>0.05),which was shown by Cox regression analysis.Ets-l plays an important role in invasion and metastasis of NSCLC.Ets-l is a useful marker for predicting the outcome for patients with NSCLC,though it is not an independent prognostic indicator.
