目的 探讨突触结合蛋白1(synaptotagmin 1,Syt1)在神经病理性疼痛小鼠前扣带回表达的变化及与痛行为的关系.方法 将20~25g的小鼠随机分为4组:假手术(Sham)+Syt1抗体(Syt1-Ab)组,Sham+对照抗体(Con-IgG)组,坐骨神经结扎(CCI)+Syt1-Ab组,CCI+Con-IgG组.在CCI第7天给每组小鼠前扣带回立体定位注射相应药物(0.1μg/0.5μl),按Hargreaves法用热辐射刺激仪测定小鼠热痛的变化,用电子测痛仪测定小鼠机械痛缩足阈值的变化,免疫印迹法检测各组小鼠前扣带回Syt1的表达.结果 免疫印迹显示神经病理性疼痛小鼠前扣带回Syt1表达明显升高,在神经病理性疼痛小鼠前扣带回立体定位注射Syt1-Ab可明显改善痛行为.结论 神经病理性疼痛小鼠前扣带回Syt1表达显著升高,在神经病理性疼痛小鼠前扣带回立体定位注射Syt1-Ab可缓解疼痛.%Objective To investigate the expression of synaptotagmin 1(Syt 1)in the anterior cingulate cortex of mice with neuropathic pain and its relationship with pain behavior.Methods A total of mice weighting 20-25 g were randomly divided into four groups:Group Sham+Syt antibody(Syt-Ab), Group Sham+Control-IgG(Con-IgG), Group chronic constriction injury(CCI)+Syt-Ab and Group CCI+Con-IgG.Corresponding drugs(0.1 μg/0.5 μl) were stereotaxically injected into the anterior cingulate cortex of mice in each group on Day 7 of CCI.The changes of ther-mal hyperalgesia were measured by the Hargreaves method.The changes of mechanical paw withdrawal threshold were de-termined by an electronic dolorimeter.The level of Syt 1 in the anterior cingulate cortex of each group was detected by Western blotting.Results The amount of Syt 1 was remarkably increased in the anterior cingulate cortex of mice with neuropathic pain.Stereotactic injection of Syt 1 Ab could significantly improve pain behavior.Conclusions The expres-sion of Syt 1 is significantly increased in the anterior cingulate cortex of mice with neuropathic pain.The pain can be re-lieved by stereotaxic injection of Syt 1 Ab in the anterior cingulate cortex.
