山药分子分类研究

         

摘要

With genome and chloroplast DNA taken as template,Dioscorea opposita 18S rDNA and chloroplast 16S rDNA fragments were amplified.A 1646bp Dioscorea opposita 18S rDNA(JF703101) and a 1351bp chloroplast 16S rDNA(JF703100) were sequenced and submitted to GenBank.Analysis of the two fragments with related sequences in GenBank shows that greater similarity lies in 18S rDNA,while chloroplast 16S rDNA has a good potential in Phylogenetic analysis of species in different family.The paper provides data for further research of Dioscorea opposita in molecular level.%提取山药基因组和叶绿体DNA作为模板,分别扩增山药核糖体18S rDNA和叶绿体核糖体16S rDNA片段.测序获得1646bp的山药核糖体18S rDNA和1351bp的山药叶绿体核糖体16S rDNA,GenBank登录号分别是JF703101和JF703100.将以上两个基因片段分别与GenBank中的一些物种的相关序列进行序列对比,结果表明,18S rDNA序列相似度较大,物种间差异较小.而叶绿体16S rDNA序列在分析不同科的物种演化关系上有较高的分辨率,为分子水平上山药资源的研究提供了资料.
