高效液相色谱法测定新疆地区甜叶菊中甜菊苷和莱鲍迪苷A的含量

摘要

目的：建立测定新疆地区甜叶菊中甜菊苷和莱鲍迪苷 A 含量的方法。方法采用高效液相色谱法，色谱柱为 Agilent·Zorbax·SB-C18（4．6 mm ×250 mm，5μm），流动相为乙腈-磷酸钠缓冲液（32∶68 v／v．pH＝2．60），流速为1．0 mL／min；检测波长为210 nm。结果甜菊苷和莱鲍迪苷 A 进样量分别在0．426～2．130μg、1．816～5．448μg 范围内与峰面积积分分值呈良好线性关系（r ＝0．9999），二者精密度、稳定性、重复性试验的 RSD≤1．50％，平均加样回收率分别为99．98％、100．06％，RSD 分别为0．86％、1．16％，测得甜叶菊中甜菊苷的含量为5．74 mg／g，莱鲍迪苷 A 的含量为10．80 mg／g。结论高效液相色谱法简便、准确、重复性好，可用于新疆地区甜叶菊中甜菊苷和莱鲍迪苷 A 的含量测定。%Objective To establish the method for the content determination of Stevioside and Rebaudioside A in Stevia rebaudiana Bertoni in Xin Jiang province.Methods HPLC method was adopted.The determi-nation was performed on Agilent·Zorbax·SB-C18 (4.6 mm×250 mm,5 μm)column with mobile phase consisted of acetonitrile-Sodium phosphate (pH=2.60 32:68 v/v)at the flow rate of 1.0mL/min.The de-termination wavelength was set at 210nm.Results The linear ranges of Stevioside and Rebaudioside-A were 0.426~2.130 μg,1.816~5.448 μg (r =0.999 9),respectively.RSDs of precision,stability and re-producibility tests were all lower than 1.50%.Average recovery were 99.98% (RSD=0.86%),100.06%(RSD= 1.16%),respectively.The average measured content of stevioside was 5.74mg/g.The average measured content of Rebaudioside A was 10.8mg/g.Conclusion The method is easy,accurate and repro-ducible,and can be used for the determination of Stevioside and Rebaudioside-A inStevia rebaudiana Ber-toni in Xin Jiang province.
