IL-1β促进食管鳞状细胞癌细胞迁移和侵袭的实验研究

摘要

Objective To investigate the effect of IL-1β on the migration and invasion of esophageal squamous cell carcinoma (ESCC) cell line.Methods ESCC cells were co-cultured with M2 TAMs as co-culture group,and the normal culture of ESCC cells were as a non co-culture group.Antibody array was used to screen the cytokines in the supernatant of the co-culture group and the non co-culture group.ESCC cells stimulated by screening of cytokine IL-1β,the ESCC cells added to IL-1β (100 ng/mL) were named as IL-1β treatment group,and PBS was added to replace IL-1β as the untreated group.After IL-1β treatment,the migration and invasion ability of ESCC cells were observed.Results The migration and invasion ability of ESCC cells were significantly increased after IL-1β treatment,which was significantly different from that of untreated group (P ＜0.05).Conclusion IL-1β significantly promotes the migration and invasion in ESCC cells.%目的 研究白细胞介素1-β(IL-1β)对食管鳞状细胞癌(ESCC)细胞迁移和侵袭的影响.方法 食管癌细胞与M2型肿瘤相关巨噬细胞(M2 TAMs)共培养,食管癌细胞加M2 TAMs作为共培养组,正常培养食管癌细胞作为非共培养组.用抗体芯片筛查共培养组与非共培养组培养液上清中表达差异的细胞因子.用筛查出的细胞因子IL-1β刺激ESCC细胞,培养基中加入IL-1β (100 ng/mL)培养48 h,作为IL-1β处理组,加入PBS代替IL-1β作为未处理组,观察IL-1β处理后ESCC细胞的迁移和侵袭能力.结果 IL-1β处理后ESCC细胞的迁移和侵袭能力均明显增加,与未处理组比较差异有统计学意义(P＜0.05).结论 IL-1β可促进ESCC细胞迁移和侵袭.
