混合培养神经元和星形胶质细胞对活性氧的应激性

         

摘要

Objective To investigate the response of neurons and astrocytesfrom Sprague Dawley rat brain against tert-buty1 hydroperoxide(tbOOH) toxicity,both separately and in cooperation, for the purpose of exploring the interrelation of the two kinds of cells. Methods Cell viability of singly cultured neurons or astrocytes from the cerebral cortex was compared by MTT reduction assay,and the morphological changes of cocultured neurons and astrocytes were observed upon tbOOH attack. Results The difference of cell viability was insignificant(P>0.05) between the neurons and astrocytes, but the damage of the neurons exceeded that of the astrocyte in terms of severity in mixed culture. Conclusion The astrocytes is very likely to possess better ability to fight tbOOH toxicity than the neurons do when they are cultured together, suggesting that the protection of neuron against active oxygen species toxicity is actually executed by the astrocyte.%目的研究混合培养脑神经元与星形神经胶质细胞对活性氧叔丁基脂氢过氧化物(tbOOH)的应激性,以探讨神经元与星形神经胶质细胞的相互作用。方法采用MTT比色法测定单独培养的大鼠大脑皮层神经元与星形神经胶质细胞在分别遭受tbOOH攻击后的细胞存活率,及用形态学方法评估二者混合培养时遭受tbOOH攻击时的细胞形态变化。结果单独培养时遭受tbOOH攻击后的细胞存活率无显著性差异(P>0.05),但二者混合培养时,神经元比星形神经胶质细胞损伤、死亡严重。结论神经元和星形神经胶质细胞共同存在时,星形神经胶质细胞较神经元对活性氧有较强的抵抗力。
