云南省部分烟区根结线虫形态和分子鉴定

         

摘要

[Objective]The present study investigated species and distribution of root-knot nematodes in tobacco grow-ing regions of Yunnan, in order to provide reference for comprehensive prevention of root-knot nematodes disease and to-bacco variety layout. [Method]During August to October, 2015, 54 samples of root-knot nematode host on tobacco crops were collected from Kunming city, Qujing city and Yuxi city in Yunnan. The species of root-knot nematodes were identified by observing perineal patterns and molecular biological testing (PCR of ITS region of ribosomal DNA and sequence charac-terized amplified region , SCAR ) . [Result]There were 24 samples belonging to Meloidogyne arenaria, accounting for 44.44%;seven M. incognita samples, accounting for 12.96%;one M. javanica sample, accounting for 1.85%. Eleven sam-ples were compound population of M. javanica and M. arenaria, accounting for 20.37%. Five were compound population of M. incognita and M. arenaria, accounting for 9.26%. Three were compound population of M. incognita and M. javanica, ac-counting for 5.56%. Three belonged to compound population of M. incognita, M. javanica and M. arenaria, accounting for 5.56%. [Conclusion]The species of root-knot nematodes in tobacco growing areas of Yunnan mainly include M. incognita, M. javanica and M. arenaria. M. arenaria is the dominant population.%[目的]明确当前云南省部分烟草种植区根结线虫的种类和分布,为烟草根结线虫病综合防治及烟草品种布局提供理论依据.[方法]2015年8~10月,在云南省主要烟草种植区昆明、曲靖和玉溪市采集54份烟草根结线虫病根样本,采用雌成虫会阴花纹形态观察及分子生物学检测(rDNA-ITS区和SCAR特异性鉴定)对病原根结线虫种类进行鉴定.[结果]采集的54份样本中有24份样本的病原为花生根结线虫(Meloidogynearenaria),占44.44%;7份样本为南方根结线虫(M.incognita),占12.96%;1份样本为爪哇根结线虫(M.javanica),占1.85%;11份样本为爪哇根结线虫和花生根结线虫的复合种群,占20.37%;5份样本为南方根结线虫和花生根结线虫的复合种群,占9.26%;3份样本为南方根结线虫和爪哇根结线虫的复合种群,占5.56%;3份样本为南方根结线虫、爪哇根结线虫和花生根结线虫3种线虫的复合种群,占5.56%.[结论]当前危害云南省部分烟草种植区(昆明、曲靖和玉溪烟区)的根结线虫种类主要有南方根结线虫、爪哇根结线虫和花生根结线虫3种,以花生根结线虫为优势种群.
