纯化的大鼠嗅鞘细胞在大鼠脊髓损伤中的修复作用

         

摘要

Objective To evaluate the effect of purified olfactory ensheathing cells in repairation of acute spinal cord injury and discuss its feasibility for therapy.Methods The olfactory ensheathing cells were cultured and purified with the olfactory bulb of Wistar neonate rats.Manufacturing transection injury model in the rat spinal cord and transplanting the puried cells with different concentration,and then changes of the spinal cord,neurologic evaluation (BBB scores) and histochemistry changes were observed.Results Immunoadsorption is a reliable way to purify olfactory ensheathing cells.When the concentration was 3 × 105 mol/L,the effect of transplantion is the best.After a continuous postoperative observation,it was proved that there was an obvious improvement in terms of motor function,and the number of nerve fiber of the transplant area has increased.Conclusion Olfactory ensheathing cells transplantation can promote the nerve regeneration and the recovery of motor function in spinal cord injuries.%目的 探讨纯化的大鼠嗅鞘细胞对损伤的脊髓的修复作用,评价其作为脊髓损伤治疗的可行性.方法 用新生Wistar大鼠嗅球做嗅鞘细胞的培养,大量繁殖并利用免疫吸附进行纯化;制造大鼠脊髓横断伤模型;将纯化的细胞以不同的浓度进行细胞移植,连续观察损伤脊髓的变化、神经功能评定(BBB评分)及其组织化学变化.结果 免疫吸附是纯化嗅鞘细胞的可靠方法,在细胞浓度为3×105 mol/L时细胞移植效果较好,术后连续观察,试验组较对照组有明显的运动功能的恢复,组织化学观察,移植区神经纤维数量增多.结论 嗅鞘细胞移植可在一定程度上促进脊髓损伤神经纤维再生和功能恢复.
