目的:探讨电针对胰岛素抵抗大鼠骨骼肌ERK、JNK蛋白表达的影响。方法:将SD雄性大鼠随机分为空白组、模型1组、模型2组、电针1组、电针2组,用高脂高糖饮食喂养法建立胰岛素抵抗大鼠模型,用葡萄糖氧化酶法检测FBG、放射免疫分析法检测FINS,并计算Homa-IR。确定造模成功后,电针组大鼠取足三里、三阴交、胃脘下俞行电针治疗,用Western blot技术检测大鼠骨骼肌中ERK、JNK蛋白的表达。结果:模型组大鼠FBG、FINS、Homa-IR较空白组显著升高(P<0.05),电针组大鼠FBG、FINS、Homa-IR较模型组显著下降(P<0.05),模型组大鼠ERK、JNK蛋白表达较空白组显著上升(P<0.05),电针组大鼠ERK、JNK蛋白表达较模型组显著下降(P<0.05)。结论:电针可降低ERK、JNK蛋白表达水平,从而改善胰岛素抵抗状态。%Objective:To investigate the influence of electroacupuncture on expression of ERK and JNK protein in the skeletal muscle of insulin resistant rat. Method:The male SD rats were randomly divided into five groups, blank group,model 1 group,model 2 group,EA 1 group and EA 2 group. To establish the insulin resistant rats model by feeding with high fat and high sugar diet. We detect FBG with the glucose oxidase method and FINS with the radioimmunoassay,and then we calculated the Homa-IR. After the successful modeling determined,the EA group were treated with electroacupuncture at Zusanli,Sanyinjiao and Weiwanxiashu. The expression of ERK and JNK protein in the skeletal muscle of rats was detected with the Western blot. Result:After electroacupuncture,the FBG, FINS and Homa-IR of each model group significantly increased more than that of blank l group(P<0.05). The expression of ERK and JNK protein of each model group significantly increased more than that of blank l group(P<0.05).The FBG, FINS, Homa-IR of each EA group significantly decreased more than that of each model group (P<0.05). The expression of ERK and JNK protein of each EA group significantly decreased more than that of each model group(P<0.05).Conclusion:The electroacupuncture could improve insulin resistant by regulating the expression level of ERK and JNK protein in the insulin resistant rat.
