目的 建立枸骨叶提取物中异鼠李素-3-O-β-D-葡萄糖苷和芒柄花素的高效液相色谱同时测定方法.方法 采用高效液相色谱法,Phenomenex C18(4.6 mm×250 mm,5 μm),甲醇-0.15%醋酸水为流动相梯度洗脱,检测波长:254 nm,流速:1 mL·min-1,柱温:30 ℃.结果 异鼠李素-3-O-β-D-葡萄糖苷和芒柄花素分别在6.97~278.8 μg·mL-1范围和2.96~118.4 μg·mL-1范围内线性关系良好,平均回收率分别为98.41%和97.43%.结论 该方法准确、可靠、重复性好,可用于枸骨叶提取物的质量评价与控制标准.%Objective To establish a HPLC method for the simultaneous determination of isorhamnetin-3-O-β-D-glucoside and formononetin in extract of leaves of Ilex cornuta.Methods The HPLC analysis was carried out on Phenomenex C18 column(4.6 mm×250 mm,5 μm) with methanol-0.15% acetic acid solution as the mobile phase for gradient elution at a flow rate of 1 mL·min-1.The detection wavelength was 254 nm and the column temperature was 30 ℃.Results A good linear relationship were found in the the range of 6.97~278.8 μg·mL-1 for isorhamnetin-3-O-β-D-glucoside,2.96~118.4 μg·mL-1 for formononetin.Their average recoveries were 99.26% and 98.75%,respectively.Conclusion The method was accurate,reliable,and can be used for the quality control and evaluation of extract of leaves of Ilex cornuta.
