内皮型一氧化氮合酶基因G894T多态性与宁夏汉族原发性高血压的相关性研究

摘要

目的 研究内皮型一氧化氮合酶( Endothelial nitric oxide synthase,eNOS)基因第7外显子G894T多态性与宁夏地区汉族原发性高血压( essential hypertension,EH)的相关性.方法 应用多聚酶链反应(Polymerase chain reaction,PCR)结合限制性片段长度多态性(Restriction fragment length poly orphism,RFLP)方法对宁夏汉族160例EH患者和176例正常血压者的eNOS基因第7外显子894位处进行基因分型,统计分析该位点不同基因型及等位基因频率在EH组和对照组中的分布.结果 EH患者组GG、GT和TT基因型分布频率为86.25％、13.13％和0.62％,正常血压组为87.50％、11.36％和1.14％,两组比较差异无统计学意义(P＞0.05)；G与T等位基因频率在EH组中分别为92.81％、7.19％,在正常血压组中分别为93.18％和6.82％,两组对比差异亦无统计学意义(P＞0.05).结论 eNOS基因第7外显子G894T多态性与宁夏汉族EH无关联性.%Objective To investigate the association between the polymorphism of endothelial nitric oxide synthesis gene G894T and essential hypertension (EH) in Han ethnicity in Ningxia Province, China. Methods The genomic DNA were extracted from the peripheral blood samples of 336 persons in Ningxia. Polymerasechain reaction (PCR) and restriction fragment length polymorphism (RFLP) methods were used to detect the G894T polymorphism of eNOS gene in Han ethnicity including 160 patients with EH and 176 normal controls. The frequencies of genotype distribution in EH and normal groups were analyzed. Results The distribution genotype frequencies of GG, GT and TT were 0. 8625,0. 13125 and 0. 00625 in hypertension groups, and those in normal groups were 0. 875,0. 113636 and 0. 011364 , respectively, which showed no significant difference(P >0.05) ; The allelic frequencies of G and T were 0.928125 ,0.071875 respectively in hypertension group and 0. 931818 ,0. 068182 in normal group. The allele frequencies of G/T were no significant differences to above data in two groups (P > 0. 05 ). Conclusion It may not be association between the G894T polymorphism of eNOS gene and essential hypertension in Han ethnicity of Ningxia.
