动脉粥样硬化患者高密度脂蛋白的抗氧化作用

     

摘要

Objective To explore antioxidant effects of paraoxonase-1(PON-1)on high-density lipoprotein(HDL)in patients with coronary heart disease(CHD)in order to provide experimental evidences for explaining molecular mechanisms of AS and prevent AS.Methods Blood samples of 199 patients with CHD were collected as CHD group including 104 patients with low level HDL-C and 95 patients with normal level HDL-C.Blood samples of 100 healthy persons were collected as control group including 22 healthy persons with low level HDL-C and 78 healthy persons with normal level HDL-C.PON-1 activity and blood lipid indexes were tested.Results PON-1 activity was lower in people with low level HDL-C than those with normal level HDL-C(P<0.01).There was no significant difference on PON-1 activity between CHD patients with normal level HDL-C and healthy persons with low level HDL-C(P >0.05).However,PON-1/apoA-I ratio was lower in CHD patients with normal level HDL-C(P<0.01).Conclusion Patients with CHD may have abnormal HDL antioxidant effects with significant decrease of PON-1 activity.PON-1/apoA-I ratio can be used as a good evaluating index for HDL antioxidant effects in patients with CHD.%目的 探讨冠心病(CHD)患者动脉粥样硬化(atherosclerosis,As)血中对氧磷酯酶-1(paraoxonase-1,PON-1)酶活性对HDL抗氧化功能的影响,为阐明As的分子机制与防治CHD提供实验依据.方法 收集199例CHD患者(CHD组)的血样,其中低HDL-C水平104例,HDL-C正常水平95例;100例正常人(对照组)的血样,其中低HDL-C水平22例,正常HDL-C水平78例.检测各组PON-1活性和血脂常规指标.结果 各组中低HDL-C水平者PON1活性均明显低于正常HDL-C水平者(P<0.01). 正常HDL-C水平的CHD患者与低HDL-C水平的正常人相比PON-1活性无明显差异(P>0.05),但前者较后者的PON-1/apoA-I比值明显降低(P<0.01).结论 CHD患者存在HDL抗氧化功能异常,其PON-1活性显著下降,PON-1/apoA-I比值可作为评价CHD患者HDL抗氧化功能的较好指标.
