Objective To study the mRNA expression of B7 - H4 in hepatocellular carcinoma( HCC) cell lines and the expression changes in BEL - 7404 under anticancer drug sorafenib. Methods The mRNA expression of B7 - H4 in ten liver cell lines was detected by reverse transcription polymerase chain reaction( RT - PCR) . With different concentrations sorafenib, we observed the change of B7 -H4 mRNA expression in BEL -7404. Results B7 - H4 mRNA was expressed partly in these liver cancer cell lines. B7 - H4 mRNA of BEL - 7404 was significantly decreased after using sorafenib. Conclusion B7 - H4 gene is closely related to tumor cell growth activity.%目的 研究B7家族的新成员B7-H4 mRNA在肝癌细胞株中的表达,以及BEL-7404在抗肿瘤药物索拉非尼作用下mRNA的表达变化.方法 采用反转录聚合酶连反应( RT-PCR) 方法检测10株肝细胞株中mRNA的表达;使用不同浓度索拉非尼杀伤BEL-7404后检测其B7-H4 mRNA的表达.结果 B7-H4 mRNA在部分肝癌细胞株中有表达;在使用索拉非尼后BEL-7404 B7-H4表达明显下降.结论 B7-H4基因与肿瘤细胞生长活性密切相关.
