银杏叶提取物对肝纤维化大鼠Ⅰ、Ⅲ型胶原及TIMP-1 mRNA、MMP-1mRNA表达的影响

         

摘要

Objective To observe the effect of Ginkgo biloba extract( GbE) on the expression of Ⅰ , Ⅲ collagen and TIMP - 1 mRNA, MMP - 1 mRNA in hepatic fibrosis rats, and to explore the mechanism of anti - fibrosis. Methods Thirty rats were divided into 3 groups (control group, model group and treatment group) , and every group had 10 rats. A group was normal control group, with no special treatment; B, C groups were made into rat liver fibrosis model. At the same time,Rats in C group were treated with Ginkgo biloba extract. The expression of Ⅰ , Ⅲ collagen were detected by immunohistochemistry and the expression of TIMP - 1 mRNA, MMP-1 mRNA were delected by RT - PCR. Results The expression of Ⅰ , Ⅲ collagen in model group and treatment group were significantly higher than that of control group (P < 0. 01 ). After treated by GbE orally, the expression of Ⅰ , Ⅲ collagen in treatment group was significantly lower than that of model group (P < 0. 01 ). The expression of TIMP - 1 mRNA, MMP - 1 mRNA in model group and treatment group were significantly increased than that of control group( P < 0. 01). The expression of TIMP - 1 mRNA in treatment group was significantly lower than that of model group (P< 0. 01) , but the expression of MMP - 1 mRNA in treatment group was significantly higher than that of model group (P <0.01). Conclusion Ginkgo biloba extract may promote the degradation of Ⅰ , Ⅲ collagen and play the role of anti -fibrosis by inhibiting the expression of TIMP - 1 and increasing the expression of MMP - 1.%目的 观察银杏叶提取物对肝纤维化大鼠Ⅰ、Ⅲ型胶原及TIMP-1 mRNA、MMP-1mRNA表达的影响,并探讨其抗肝纤维化的作用机制.方法 30只大鼠随机分为3组(对照组,模型组和治疗组),每组各10只.A组为正常对照组,不做特殊处理,B、C两组制备大鼠肝纤维化模型,同时C组给予银杏叶提取物治疗;采用免疫组化和RT-PCR方法分别检测3组大鼠肝脏组织Ⅰ、Ⅲ型胶原及TIMP-1 mRNA、MMP-1mRNA的表达情况.结果 模型组及治疗组肝脏组织Ⅰ、Ⅲ型胶原表达水平明显高于对照组(P<0.01),经银杏叶提取物(GbE)灌胃处理后,治疗组肝脏组织Ⅰ、Ⅲ型胶原表达水平明显低于模型组(P<0.01);模型组及治疗组肝脏组织TIMP-1 mRNA、MMP-1 mRNA表达水平较对照组均显著增高(P<0.01),且治疗组TIMP-1mRNA表达水平明显低于模型组(P<0.01),治疗组MMP-1 mRNA表达水平明显高于模型组(P<0.01).结论 银杏叶提取物可能通过抑制TIMP-1表达和提高MMP-1表达而促进Ⅰ、Ⅲ型胶原降解并发挥抗肝纤维化的作用.
