水凝胶作为载体的口服胰岛素制剂的药代动力学及药效学研究

         

摘要

目的 对以载糖蛋白-水凝胶为载体的口服胰岛素制剂进行药代动力学及药效学评价.方法 建立糖尿病大鼠模型,实验分空白组、皮下给药组(2 U/kg),以及两个口服胶囊组,口服胶囊组给药剂量分别是50 U/kg和100 U/kg(制剂胰岛素含量依次为200和800 U/g).建立酶联免疫法(ELISA)对胰岛素的吸收进行检测,并采用血糖仪监测血糖水平的变化情况.结果 皮下组给药后血糖变化及胰岛素吸收情况均较好,口服组给药后未检测到胰岛素的吸收,血糖基本维持稳定状态.结论 以载糖蛋白-水凝胶作为载体的口服胶囊没有起到降血糖作用,整个实验的设计与思路为从事相关研究的学者提供了参考,有利于对水凝胶或其他生物材料作进一步的修饰改善.%Objective To evaluate the pharmalokinetics-pharmacodynamics properties of an oral insulin delivery system using a special hydrogel conjuncted with glycoprotein as carrier. Methods Streptozotocin-induced diabetic rats were randomized to four groups; 2 U/kg subcutaneous administration group, SOU/kg and lOOU/kg oral administration groups (200U/g and 800U/g insulin content, respectively) and control group with empty capsules. At different time points we detected the blood glucose with blood glucose monitoring (Omron, USA) and the concentration of insulin by ELISA kit Results The subcutaneous administration group had a good insulin absorption and blood glucose change. However, in the control group and two oral administration groups insulin absorption was not dectected and the blood glucose level ratio kept stable. Conclusion The technique of oral insulin hydrogel preparation needs further improvement
