高效液相色谱法测定不同保存时间温莪术中四甲基吡嗪的含量

摘要

Objective To establish a determining method for tetramethylpyrazine in Curcuma aromatic Salisb by high-perfomance liquid chromatography. Methods Tetramethylpyrazine was extracted from Curcuma aromatic Salisb by methanol. Kromasil C18(250 mm×4.6 mm,5μm)was used as stationary phase,methanol: 0.8 % sodium acetate solution(42∶58)was used as mobile phase,the flow rate was 1 ml/min and the wavelength of detection was at 282 nm. Results The calibration curve showed good linearity within the concentration range of 2~20 ng,r=0.9999,and the average recovery was 99.56%,RSD=0.65%. Conclusion The method is simple,quick and suitable for the determination of tetramethylpyrazine in Curcuma aromatic Salisb. No literature home and abroad has yet reported quality control of Curcuma aromatic Salisb by tetramethyl pyrazine.%目的：建立不同保存时间中药温莪术中四甲基吡嗪的含量测定方法，以控制温莪术质量；方法用甲醇提取，采用 Kromasil C18（250 mm×4.6 mm，5μm）色谱柱；以甲醇：0.8%醋酸钠溶液（42∶58）为流动相，流速：1 ml/min，检测波长：282 nm。以外标法测定温莪术中四甲基吡嗪的含量；结果四甲基吡嗪在2～20 ng 范围内呈良好的线性关系，r =0.9999；平均回收率为99.56%，RSD =0.65%。结论该方法操作简便、快速、有良好的准确度，目前国内外文献还未见报道以四甲基吡嗪控制温莪术质量，所以本法可作为中药温莪术中四甲基吡嗪含量的测定方法，以控制温莪术质量，或为其质量控制提供参考。
