胃癌组织中TS及TUBB3表达量与癌细胞侵袭性生长的相关性研究

摘要

目的:探讨胃癌组织中TS及TUBB3表达量与癌细胞侵袭性生长的相关性.方法:2016年7月~2017年12月间在延安大学附属医院确诊并接受胃癌根治术的原发性胃癌患者98例,术中留取胃癌组织、癌旁组织,采用荧光定量 PCR法检测其中 T S、T UBB3基因及侵袭相关基因的表达量.采用Pearson检验评估胃癌组织中TS、TUBB3表达量与侵袭相关基因表达量的相关关系.结果:胃癌组织中TS、TUBB3 mRNA表达量高于癌旁组织(P< 0.05);促侵袭基因 Ezrin、Tiam1、UHRF1、MTA1 mRNA的表达量高于癌旁组织(P<0.05),抗侵袭基因PTEN、NDRG2、ARID1A、LKB1 mRNA的表达量低于癌旁组织(P< 0.05).Pearson 检验发现,胃癌组织中 TS、TUBB3表达量与促侵袭基因Ezrin、Tiam1、UHRF1、MTA1 mRNA的表达量呈正相关(P<0.05);与抗侵袭基因PTEN、NDRG2、ARID1A、LKB1 mRNA的表达量呈负相关(P<0.05).结论:胃癌组织中 TS、TUBB3基因异常高表达,具体表达量与癌细胞侵袭活性直接相关.%Objective:To investigate the correlation of TS and TUBB3 expression in gastric cancer tissue with the inva-sive growth of cancer cells.Method:A total of 98 patients with primary gastric cancer who were diagnosed and underwent radi-cal operation for gastric cancer in Yan'an University Affiliated Hospital between July 2016 and December 2017 were selected, gastric cancer tissue and adjacent tissue were collected during operation,and fluorescence quantitative PCR method was adopted to detect the expression of TS,TUBB3 and invasion-related genes.Pearson test was used to evaluate the correlation of TS and TUBB3 expression with invasion-related gene expression in gastric cancer tissue.Results:TS and TUBB3 mRNA expression in gastric cancer tissue were higher than those in adjacent tissue(P< 0.05);pro-invasion genes Ezrin,Tiam1,UHRF1 and MTA1 mRNA expression were higher than those in adjacent tissue(P<0.05)whereas anti-invasion genes PTEN,NDRG2,ARID1A and LKB1 mRNA expression were lower than those in adjacent tissue(P<0.05).Pearson test showed that the TS and TUBB3 expression in gastric cancer tissue were positively correlated with the pro-invasion genes Ezrin,Tiam1,UHRF1 and MTA1 mRNA expression(P<0.05),and negatively correlated with anti-invasion genes PTEN,NDRG2,ARID1A and LKB1 mRNA expression(P<0.05).Conclusions:The TS and TUBB3 gene expression are abnormally high in gastric cancer tissue and directly correlated with the invasion activity of cancer cells.
