Objective:To investigate the expression of calreticulin (CALR) in human gingival fibroblasts (HGFs) at different time under the action of mechanical pressure.Methods:HGFs were cultured by tissue block culture method.The cells were seeded on the PLGA scaffold,and then subjected to 25 g/cm2 loading force for 0 h (control),6 h,24 h,48 h and 72 h,respectively.The expressions of CALR mRNA and protein were detected by fluorescence quantitative PCR (qPCR) and western blotting,respectively.Results:The mRNA and protein expressions of CALR were up-regulated with the increasing of loading duration,and reached the highest level at 24 h,then decreased gradually.Conclusion:Mechanical compressive stress could up-regulate CALR expression in HGFs.The CALR level increased with prolonged duration of loading force,and reached the peak then decreased.%目的:探究机械压应力作用下不同时间人牙龈成纤维细胞(HGFs)钙网蛋白(CALR)的表达.方法:采用组织块培养法培养HGFs,将细胞接种在PLGA膜上,采用重物加载方式对其施加25 g/cm2的压应力,按加力时间不同分为0h组(对照组)、6h组、24 h组、48 h组、72 h组,分别采用荧光定量PCR法(qPCR)和western blotting法检测各组CALR基因和蛋白表达量.结果:随着加力时间的延长,CALR基因和蛋白表达量升高,在24 h时升至最高,随后逐渐降低.结论:机械压应力可使HGFs中CALR表达上调,且随着加力时间的延长CALR表达水平增高,到达峰值后下降.
