黄芪多糖对脑缺血再灌注大鼠脑组织Caspase-3表达及形态学的影响

         

摘要

In order to study the effects of Astragalus polysaccharides on Caspase-3 expression and mor-phology in rats brain tissue after cerebral ischemia reperfusion,72 SPF level Wistar rats were randomly di-vided into 6 groups,sham operation group,model group,positive control group,Astragalus polysaccharides high dose group,Astragalus polysaccharides middle dose group and Astragalus polysaccharides low dose group,1 2 rats in each group.With modified thread embolism method,the cerebral ischemia reperfusion models of rats were established.Each drug group was respectively given corresponding medicine for 1 5 days,and was detected after put to death.The changes of rats brain tissue in each group were observed by HE staining,and the expression level of Caspase-3 protein in rats brain tissues were detected with immuno-histochemical method.The results showed that Caspase-3 expression was weak in artificial surgery group, which was much stronger in model group (P<0.01).Compared with model group,Caspase-3 expression a-mount of other groups were obviously less (P<0.05,P<0.01),particularly in Astragalus polysaccharides high dose group.The morphological changes in model group appeared obvious apoptosis,neurons disap-pear,mesenchyme loosen,karyopyknosis and chromatin condensation,nerve cell apoptosis of other groups were less worse than that of model group,especially obvious in Astragalus polysaccharides high dose group.The study showed that Astragalus polysaccharide could significantly inhibited nerve cell apoptosis, this should be realized by reducing the expression of Caspase-3,and the inhibition effect was dose-depend-ent.%为了探讨黄芪多糖(Astragalus polysaccharide,APS)对大鼠脑缺血再灌注损伤脑组织 Caspase-3表达及形态学的影响.选取 SPF级 Wistar大鼠72只,随机分为假手术组、模型对照组、阳性药物对照组、黄芪高剂量组、黄芪中剂量组、黄芪低剂量组,每组12只;采用改良线栓法建立大鼠脑缺血再灌注模型,各药物组分别给予相应药物15 d,后处死检测,HE染色观察各组大鼠脑组织形态学变化,免疫组化法检测各组大鼠脑组织中 Caspase-3蛋白的表达水平.结果表明:假手术组大鼠Caspase-3表达较弱,模型组较假手术组明显增多(P<0.01);与模型组比较,其余各组大鼠的Caspase-3表达均有显著减少(P<0.05),以黄芪多糖高剂量组大鼠尤为明显.形态学试验结果表明模型组大鼠可见神经元大片消失,间质疏松,残余细胞胞核固缩、染色质凝聚,其余各组神经元细胞发生凋亡的程度较模型组轻,以黄芪高剂量组效果明显.说明黄芪多糖能显著抑制脑缺血再灌注损伤后的神经细胞凋亡,其作用可能是通过降低Caspase-3的表达实现的,且作用效果呈剂量依赖性.
