Objective To compare the different methods of preparation of type 1 pneumococcal polysaccharide - protein conjugate vaccine biochemical and immunological properties. Methods amine reduction and cyanogen bromide activation method was used to conjugate prepared and take biochemical and immunological detection methods for its detection. The results relative to the amine reduction conjugates produced using cyanogen bromide activation method combined with high polymer content and higher binding rate, while the immunized mice produced antibodies increased very significantly. Conclusion reduction relative to the amine conjugates produced using cyanogen bromide activation method for type 1 pneumococcal polysaccharide - protein conjugate vaccine prepared to excellent.%目的:比较不同方法制备的1型肺炎球菌多糖-蛋白结合疫苗生化及免疫学特性。方法采用胺还原法以及溴化氰活化法分别对结合物进行制备,并采取生化以及免疫学检测的方式对其进行检测。结果相对于胺还原法制造出来的结合物,利用溴化氰活化法具有较高的高分子结合物含量以及较高的结合率,与此同时,免疫小鼠产生的抗体提高的也十分明显。结论相对于胺还原法制造出来的结合物,利用溴化氰活化法对1型肺炎球菌多糖-蛋白结合疫苗进行制备要优。
