丹红注射液激活AMPK保护心肌缺血再灌注损伤模型大鼠的实验研究

         

摘要

目的 探讨丹红注射液激活AMPK保护缺血再灌注大鼠心肌的相关机制.方法 将健康SD大鼠36只随机分为对照组、对照加丹红组、缺血再灌注组和缺血再灌注加丹红组.分别取各组大鼠心肌组织,血清.1)观察各组大鼠血清中TnT、LDH变化;2)用HBFP染色法将各组大鼠心肌组织进行染色,观察其坏死变化情况;3)采用投射电镜观察心肌组织中线粒体形态及结构变化;4)提取心肌组织蛋白,测定各组p-AMPKα、AMPKα的表达.结果 丹红注射液可以降低大鼠心肌缺血再灌注的血清TnT、LDH水平、减少心肌坏死、减轻心肌细胞线粒体损伤、上调心肌AMPK的磷酸化水平.结论 丹红注射液激活AMPK减轻大鼠心肌缺血再灌注损伤及线粒体损伤.%Objective:To study the protective effects of Danhong Injection on myocardial in ischemia reperfusion injury by activating of AMPK.Methods:36 healthy SD rats were randomly divided into the control group,Danhong group,ischemia reperfusion group,ischemia reperfusion and Danhong group.Myocardial tissue and serum were taken from each group.1)The level of TnT and LDH was observed.2)Morphological changes were observed through HBFP staining.3)Ultrastructure morphological changes were observed through transmission electron microscopy.4)The expression of p-AMPK and AMPK alpha was detected through western blotting.Resuits:Danhong Injection can decrease the levels of serum TnT and LDH,reduce myocardial necrosis,relieve mitochondrial damage and up-regulate the phosphorylation of AMPK in myocardial ischemia reperfusion rats.Conclusion:Danhong Injection reduces myocardial ischemia reperfusion injury and mitochondrial injury through the activation of AMPK.
