化湿降浊方对高尿酸血症模型大鼠血尿酸、尿尿酸水平及肾功能的影响

     

摘要

Objective:To observe the effect of Huashi Jiangzhuo decoction on hyperuricemia rat model and do the preliminary study of Huashi Jiangzhuo decoction to improve the uric acid metabolism.Methods:Hyperuricemia rat model was replicated by intragastric administration of hypoxanthine combined with potassium oxonate injection.Benzene bromine Malone was taken as positive drug control,the effect of Huashi Jiangzhuo decoction (1.2 g/mL) on serum uric acid (UA),urine UA,serum xanthine oxidase (XOD),blood urea nitrogen (BUN),crea tinine(Scr) was oberved.Results:Compared with the control group,the model group significantly increased blood UA and serum XOD(P< 0.01).Compared with the control group,traditional medicine group and benzene bromine Malone group effectively promoted urinary UA excretion inhibited the activity of serum XOD in order to reduce the blood UA,the difference was significant(all P <0.01) traditional medicine group and benzene bromine Malone group decreased the abnormal serum Scr and content of BUN and the difference was significant(all P< 0.01);the traditional medicine group did it better than the benzene bromine Malone group on decreasing the abnormal serum Scr,the difference was significant (all P < 0.01).Conclusion:Intragastric administration of hypoxanthine combined with potassium oxonate by intraperitoneal injection can replicate hyperuricemia model rats successfully;Huashi Jiangzhuo decoction can obviously promote urinary UA excretion,inhibit the activity of serum XOD,reduce blood UA to protect the renal function,thus is worthy of further research and promotion.Gu Ning is the anthor of communication.%目的 观察化湿降浊方对高尿酸血症模型大鼠血尿酸、尿尿酸及肾功能的影响.方法 采用灌胃次黄嘌呤联合氧嗪酸钾腹腔注射方法复制高尿酸血症大鼠模型.以苯溴马隆为阳性药物对照,研究化湿降浊方(1.2 g/mL)对高尿酸血症大鼠血尿酸(UA)、尿UA、血清黄嘌呤氧化酶(XOD)、尿素氮(BUN)、肌酐(Scr)的影响.结果 与空白组对比,模型组大鼠血UA、血清XOD的活性显著升高(均P<0.01),尿UA水平下降(P<0.01).与模型组相比,中药组与苯溴马隆组都能不同程度地促进大鼠尿UA排泄、抑制血清XOD的活性以达到降低血UA的作用,其差异具有统计学意义(均P<0.01).中药组与苯溴马隆组都能不同程度降低模型大鼠异常的血清SCr、BUN水平,与模型组大鼠比较,差异均有统计学意义(均P< 0.01),且中药组较苯溴马隆组降低异常的血清SCr作用更明显(P<0.01).结论 灌胃次黄嘌呤联合氧嗪酸钾腹腔注射可以成功复制高尿酸血症模型大鼠;化湿降浊方可明显促进模型大鼠尿UA的排泄,具有抑制血清XOD的活性,从而达到降低血UA目的,并能一定程度改善异常的肾功能.
