X型胶原在膝骨关节炎软骨中的表达

摘要

目的 观察膝骨关节炎患者关节软骨中X型胶原的表达特征,探讨X型胶原在骨关节炎疾病进展中的作用.方法 取18例膝骨关节炎患者和13例正常对照者的关节软骨.以Northern杂交、RT - PCR和Western杂交,分别检测各组样品X型胶原mRNA和蛋白水平的表达情况.结果 Northern杂交与RT-PCR的结果均显示膝骨关节炎的X型胶原表达有显著升高.Western杂交发现膝骨关节炎患者软骨基质中X型胶原的含量上升(P＜0.01).但分析mRNA和蛋白水平的变化趋势,未能发现它们之间存在明确的相关性(P＞0.05).结论 膝骨关节炎患者的X型胶原表达明显增加,这可能促使基质组分发生改变,从而加速了骨关节炎的进展.%Objective To observe the expression of X - type collagen in articular cartilage of patients with knee osteoarthritis (OA) and the role of it in the progression of OA. Methods The articular cartilage from 18 OA patients and 13 healthy subjects were included in the study. The synthesis of X- type collagen was measured by Western Blotting,Northern Blotting and RT - PCR. Results At mRNA level,Northern blotting and RT - PCR both revealed OA articular chondro-cytes with higher X- type collagen expression than controls. Western Blotting test showed that the synthesis and deposition of X - type collagen in OA cartilage matrix increased significantly compared with the healthy age-matched controls (P<0.01). But there was no correlation between mRNA and protein synthesis of type - X collagen( P >0.05). Conclusion The X - type collagen is highly expressed in OA patients,which probably contributes to the disorder of pericellular matrix leading to the acceleration of osteoarthritis.
